If your sequence contains more than 14 nucleotides, the equation is Tm=64.9+41[(G+C - 16.4) / total in sequence]. If your sequence contains less than 14 nucleotides, the equation is Tm=2(A+T)+4(G+C). In the equation in your video, the answer should be 46.77. The equation would look like Tm=64.9+41[(8-16.4)/19] = 46.77
