Mam can we directly file a PCT application without filing in national office (ex. Indian office)

You can file PCT application directly without filing in India after taking foreign filing permission from India

Mam do we need to file complete specification before filing pct,also can we enter again in India after filing pct.

No need to file complete specification before filing PCT, if you have filed a provisional application. But if you do not file either provisional or complete then you need to take permission as per section 39 of the Indian Patent Act. Yes, you can enter India again but in that case, Indian Patent Office will ask which application you want to examine.

It depends on you. If you do not file chapter II or supplementary search request then your PCT process will complete in 18 months from priority and you will have 30 or 31 months from priority to file the National phase applications.
