This really shows Regina's character development, in season one, she would have rejoiced at Snow having to kill her true love and even more at the thought of her death, and this time she's clearly upset at having them to separate them and doesn't want to split Snow's heart in case it doesn't work. She's acting like Snow's stepmother and the interaction between them with Snow on her knees and Regina going down at her level mirrors when they first met. Regina's face when it works is perfect first she's shocked, then she's happy and relieved. These actors are wonderful
And the fact that Regina was hesitant to do it, fearing that Snow will die, and looked happy and relieved that it worked. This shows once and for all that she is no longer The Evil Queen

Because it was the reason for everything happened, Snow gave up Emma -> Emma met Neal -> Henry was born -> regina adopted him. If she has to choose Henry or Daniel, she would choose Henry.

Snow just knew she couldn't live without charming so she had to sacrifice everything for that one last chance. While emma knew she could move on from neal. He wasn't her true love.
