Nope, doesn't matter. The string is basically just used as a straight edge between the 1st fret and the 17th (or whatever fret meets the body on your guitar.). It makes more sense visually, but you're basically measuring from the bottom of the string, that's why you zero it on top and push the string down to the fret. The distance between the bottom of the string to the top of the 7th fret is the distance you're calculating. I hope that makes sense!
