Whats the best use for exp boosters aside from filling up 6th Job Stones? Should I use them on my main or a Mule I wanna get to 210 or 220?

If youre talkimg about vip booster (red potion thing that boosts spawn for 100s) use it on main first and consistently. You can only use 10 of them a day per chatacter. If you have a surplus then use them on other characters
