  • Metformin: Mechanism of action
    Metformin is an oral anti-diabetic drug, which belongs to biguanide class. It is the only drug available in this class.
    Still now it’s difficult to find a full explanation of the mechanism of action of metformin.
    However, the primary effect of metformin is to activate the enzyme adenosine monophosphate activated protein kinase which is also called AMPK.

    Metformin activates AMPK enzyme in liver which inhibits the gluconeogenesis. The inhibition of gluconeogenesis leads to decrease glucose production.
    Patient with type 2 diabetes have considerably less fasting glycemia as well as lower post prandial hyperglycemia after administration of metformin. Also hypoglycemia is rare during metformin therapy. That’s why it is more appropriately termed as euglycemic agent.
    There are other mechanisms also, like:
    It increases the insulin sensitivity
    Increases the fatty acid oxidation
    Decreases the intestinal glucose absorption

    went from 90 to 120 kgs in 5 years. result? insulin tolerance that creates constant hunger even after 1-2 hours of eating properly. my doctor started 1000mg/day on metmorfin and after a few weeks of getting used to it I'm finally seeing results. now it takes at least 6-7 hours for me to feel hunger after a meal. lost 2 kgs already. gotta lose 20 more and I'm golden.

    Ive been taking this for roughly two months now. I have not noticed much decrease in hunger. Ive noticed a slight uptake in appetite. I eat and feel hungry a few hour later as if i had not eaten anything.

      What are you eating? It might have caused a sugar spike then crash which makes you hungry. Also it is advised often to eat smaller more frequent meals. Exercise is always helpful, too. Not just for weightloss but for your body in general and your mind. Of course double check my advise with your doctor because I'm not a doctor at all, I just have learned some stuff along the way.

      change your diat to fat mostly, eat IF only, stay healthy !

      He explains this under “other mechanisms”.
      - Increased insulin sensitivity
      -Increased Fatty Acid Oxidation
      - Decreases intestinal glucose absorption
      - Delays gastric emptying.
      You will more readily burn fat.
      You will have more stable blood sugar levels.
      You will absorb less of the potential glucose consumed.
      You will feel fuller for longer, likely increasing time between meals.

    It does indeed reduce appetite. I've stopped eating in between meals.

      @Thru the eyes of God right after breakfast and and after dinner. Do not eat empty stomach. You'll see effects after couple of doses.
