    15:15 It takes 5 half lives to reach steady state, but Half Life 3 still isn't 😞 here
    I don't think anyone reads anything that's written here. But here's for the metadata: this video is explicitly about adrenergic receptors. More specifically, the Alpha subtype, and even more specifically, Alpha 2 Adrenergic Receptors. This thing throws a massive curveball to learners in all medical disciplines because Alpha 2 agonism / antagonism are opposite of Alpha 1. The topic of these receptors can be a few weeks long in a Pharm course in Medical and Pharmacy schools, and that course would be dedicated specifically to cardiac pharmacology, so this is as much I can cover in an 18 minute video. There might be people who will go "nooooo u cant just say that about my tree bark supplement" but the point is and has always been to teach about the pharmacology, its mechanism and how the setting of excess changes the pharmacokinetics-- the agent serves as a means for me to teach that because it has that activity. If it didn't have that activity, I'd talk about something else that does in this setting. It's a fascinating molecule, like all natural products. If you don't need to ingest it, don't ingest it, but I trust you are well in your best capacity to make decisions for yourself.
    These cases are patients who I, or my colleagues have seen. They are de-identified and many instances have been presented in more depth in an academic setting. These videos are not individual medical advice and are for general educational purposes only. I do not give medical advice over the internet.

    Shoutout to one of the most useful talents of the animal kingdom: barfing. It sounds like he vomited profusely very shortly after ingesting the compound, so it's very possible that he managed to prevent way more than half of it from getting into his bloodstream. And he probably needed to be conscious for that to happen, so he did a good job by not passing out immediately. Some toxins can directly cause someone to vomit. But very often, the reason we vomit is because when we are conscious, we sometimes assess an altered state. Dizziness, headache, numbness, hallucinations, a sense of dread. The body's instincts say, "Maybe we ate something that is poisonous and it's affecting our brain. Let's throw up!" But the brain doesn't know it's in an altered state if it's in standby mode at the time. If you're not conscious, you won't know if you're dizzy, hallucinating, etc.

      Vomiting is a big win LOL. But yeah I've felt this, and I'm sure most people have also felt this. I ate some pizza one night and a combination of anxiety and a migraine caused me to go into a panic attack as I felt that sense of doom lurking over me. And, because of that doom, my body definitely felt like that pizza I ate had to come back up, even if it wasn't actually bad.

    • @tom_something
      @tom_something 7 місяців тому +234

      @@SneakingMOUSE It's even been suggested that the reason seeing someone else vomit or smelling someone else's vomit can cause us to throw up is because we have evolved as social eaters who share food. So if my cousin over there is throwing up, maybe we've both eaten something toxic, and it just hasn't kicked in yet for me.

    • @SneakingMOUSE
      @SneakingMOUSE 7 місяців тому +40

      @@tom_something Yeah, that's interesting. I enjoy learning more about the brain. I never put much thought into why we vomit, it's always been a simple answer to me. But, there's much more to it of course! Evolution has taken its course, but we still show an incredible amount of traits from our long distant ancestors.

      Yeah, a lot of Yohimbine products (yohim*be* is the tree whose bark contains many alkaloids, yohim*bine* is a single alkaloid that was isolated from the bark-) fall into this legal grey area. If they’re not claiming to treat anything the FDA really can’t do much.
      So you never know 100% what you’re getting and how much.
      I think humans rarely ever get it in drug form…we give it to animals to reverse xylazine (Anased/Rompun) anesthesia but all of those are pretty much animal only.

    I was actually researching yohimbine a while ago as an addon to my pre-workout for the same reason the patient in the video took it, but after reading the mechanism of action, I saw that it's contraindicated with clonidine and decided it wasn't worth it. I take clonidine for stimulant induced motor tics, and I can say, without a doubt, I'd rather have stubborn fat without tics than to have tics without stubborn fat. Also, this video is amazing, even for those who can hardly understand how the body works, and helped me understand what's happening when two opposing pharmacological substances are combined (in this case, clonidine administered as an antidote[?] for yohimbine overdose.)

      I've used preworkout with yohimbine and had to give it up immediately because it gave me the shakes (like the low overdose symptoms at 11:31 now that I see it) and even worse the shits,then switched like ADHD meds and made me tired. So I'd take the preworkout and even down at the caffeine level of a cup of coffee instead of going hard for a workout I'd be on the toilet shaking and dropping outright sludge until I went back to bed. Whole day ruined.

      @@ffwast Stimulants effect the stomach. Some people get the runs when they do a big amount of coke or amphetamines. And yeah the shakes weren't good and it reminded me of an uncomfortable level of caffeine. Caffeine overdose is still the most uncomfortable I have been from a drug (I didn't mean to do a bunch of caffeine but my barista friend kind of screwed me over unknowingly).
      Also funny how ADHD meds make people with actual ADHD tired and make everyone else wired.

      Yes, my regular doctor prescribed those early on. Not sure if that's normal... 🤔 But they do calm people down, myself included. I understand many people use them before high-stress situations in order to keep calmer, for instance orchestral musicians are known to use them before tests. I gave some to my niece who had failed her driving exam thrice, and they helped her pass the fourth time.
      In my case, they only helped a little. I can only eat two a day, or I start to get really dizzy. And my anxiety is pretty hefty, so they really didn't help much on their own. This was before I started with my psychiatrist (there's at least a waiting time of half a year for psychiatrists here in Denmark, as there aren't enough around to keep up with demand). Eventually, we discovered Pregabalin, which is very effective, but I've kept taking the beta blockers, as they help a little, and my blood pressure has also gotten high as a result of my weight increasing a lot over the past couple of years.
      Edit: I should note that my ADHD was also undiagnosed at this point, so I was completely unmedicated then.

    That there is a connection between opioid effects and Beta-2 is heavily indicated. We use Clonidine sometimes in cases where there is opioid dependancy that renders normal pain management difficult. Clonidin enhances effects of opiods but has side effects that needs to be monitored but can be used in hospital. The clonidine basically allows you to relax *just* enough to be able to fall asleep for a little bit. In July I committed to competing as a bodybuilder (natural), and I have since immersed myself in the culture. There are a lot of questionable practices, even among the natural competitors. Some of the supplements they take to lean out are extremely dangerous, and they're completely ignorant to the risks. Considering the effects they're going for, it's a dangerous game they're playing. Messing around with stimulants and diuretics can go south very quickly with dire results. I personally worked with Xylazine for over 2 years, assuring quality of product every step of the production. It's a helluva drug, I mean, when I worked with it, even with a respirator, gloves and standard PPE, I still would be feeling off at the end of the workday. I don’t use illegal drugs anymore, but some of my friends still do + I’m involved in harm reduction circles, and there are confirmations in my area (Pittsburgh) that Xylazine is in the local supply. I stopped using four years ago when I found out my dealer was faking negative fent test strips for her “Xanax” and “ketamine.” I almost died from my usual “Xanax” dose; tested it myself with some free test strips from a local clinic and found out it was fent.
      But we don’t have Xylazine test strips here yet and it’s terrifying. Your pill might be fent negative but be tainted with Xylazine and kill you before you have a chance to notice something’s wrong. It’s scary, and we in the harm reduction crowd feel helpless.

    When I make my own gun cotton (nitrocellulose) or even black powder for my old-timer muskets, I am working with powders and acids. All of them look pretty much the same. I am no chemist, failed college chemistry three times before transferring to physics, but even I know that you do NOTHING without checking the label three times over and then give it another look.
    Granted, I am not making a drink to ingest, I am making (legal) explosives to use as gunpowder for my cannon and firearms - HEY, ATF, propellant, NOT an explosive charge!
    I cannot understand how people inadvertently mix the wrong chemical. You are supposed to have every chemical in a labeled container with each label clearly visible from at least an arm's length away.
    I simply do not understand people who mix their own... whatever, and do not pay close attention to what is being scooped, poured or added.
    In my case, putting the wrong chemical in the mix in the wrong order equals a blast and fire, so if I screw up, I know it immediately. Because I only make small portions at a time, at worst, I will be missing fingers or have shrapnel from the container pierce my heart and kill me. At best, I will have ruined all my chemicals and workbench.
    ... but something you consume? I don't care if every chemical is the same color and texture, you put a permanent label on it so you can read it long before you reach for it... and in Sharpie, you write what it is all around the container so it is visible no matter how the container is rotated.
    At every angle, "Protein Powder"... "Sugar"... "Caffein" should be legible and then the official label on one side... even on the side of the label (with the fine print on it) use that Sharpie to write what it is.
    IT IS COMMON SENSE!
    Don't get complacent!

    As an individual who has ADHD and depression. I can say that too much dosage of an ADHD medication can be a problem because then you will have trouble falling asleep. I currently take Vyvanse and clonidine to actually get me to sleep. Otherwise I would not be able to get to sleep. ( before the clonidine I was an insomniac maybe getting 3-4 hours sleep and always being cranky) This is a problem in and of itself because I know that taking a stimulant and depressant everyday is probably not good on my body. So I ended up reducing my Vyvanse dosage from 40 mg to 20 mg. It's easier to fall asleep now. But for about 10 years at 40 mg. I was dealing with the heart rate side effect. (Imagine running and exercising but that was my heart rate 24/7 with this medication and I thought that side effect was normal. )But yeah. Don't go as long as I did before I came to my realization that my dosage was a bit too high. I'm just hoping I have no heart problems as I reach 60 or something.

      i dont think you will need to worry, the range of elevation is still within what a healthy body can normally handle, if you where experiencing severe tachycardia that would be one thing, but think about it this way, tweakers who run around for sometimes 20 or more years before quitting often dont show heart damage, and those are people ingesting sometimes GRAMS of methamphetamine which is a bit over twice as potent as the dextroamphetamine Vyvanse breaks down into.
      i wouldent worry about it too much as long as your keeping you dont have preexisting conditions and keep up on checkups and monitoring

      I had a microbiology lecturer in the mid 80s who managed to put us off just about every food imaginable with her stories of food poisoning she used to illustrate her lectures. In the end, you came to realise that so long as you treated the food appropriately, cooked, and stored it correctly, you had no issues.
      However, if you cooked some lobster and shrimp in your hotel room in Florida, wrapped it in a plastic bag, then took 3 days, driving back to Michigan then made a seafood dish for your family and wondered how you gave them all Cholera....well.... Her stories stuck in our minds, and we still talk about them today. She was one of the leading microbiology investigators for the Health Department back then and specialised in Salmonella.
