You pick two restriction sites based upon the enzymes that you want to use and delete the nucleotides between them. Then you go to NCBI, PFGC5941, the location of the bar gene, the gene you want to study. You cut the bar sequence out of NCBI and paste it between the two restriction sites. Then the primers are picked to include the restriction sites. You always have to be careful to have the plus strand, minus strand correct. Is that right?
