the purple impurity may be related to your carbonate; I recently carried out an unrelated reaction (formaldoxime from formaldehyde + hydroxylamine, using sodium carbonate to get basic conditions) and got a really deep purple colour at first, that faded on heating strangely, when it cooled down again it became an orange/brown colour

For the diphenhydramine synthesis don’t use α-bromodiphenylmethane. I couldn’t get it to work. IIRC, Thyzoid tried using that as well, and got the same result I did. I think you have to use benzhydrol. There’s also a nice easy procedure for making benzhydrol in Vogel (reduction of benzophenone with zinc and NaOH in ethanol). N,N-dimethylaminoethanol is also sold as a supplement as the bitartrate salt. It’s called DMAE bitartrate. I found that heating it in methanol with NaOH, filtering to remove the sodium tartrate, and distilling off the methanol was a decent way of isolating freebase DMAE. Can’t wait to see what you come up with!
I actually found a different way!! First I’ll reduce the benzophenone with sodium borohydride, then there is a patent that used tosic acid, toluene, and DMAE to make it. Really cool stuff! Also I literally just got done with doing DMAE synth haha. Though That Chemist showed me a way of using sodium ethoxide in ethanol to get the DMAE freebase rather NaOH. I got some stuff, but I need to fractionally distill since it likely formed a azeotrope and distilled over around 90C. Hopefully that fixes it.
@@chemdelic Awesome! Did you use Eschweiler-Clarke methylation of 2-aminoethanol to prepare DMAE? If you have any you can try adding 4A molecular sieves before distillation to remove water, ethanol, and methanol if there's any in there. That might help because if it came over as an azeotrope then you're probably not going to be able to purify by distillation alone. I suggest giving the zinc reduction of benzophenone a try instead given that NaBH4 is more expensive, and harder to get than zinc. The procedure is listed under Exp. 5.30 on page 523 of Vogel's Textbook of Practical Organic Chemistry. It takes about three hours, and gives good yields. They claim a crude yield of about 96% (m.p. of 65 C, and 68 C after recrystallization) which is about what I got as well. Plus it forms a deep blue color after adding the zinc which is nice. If you don't have a copy of Vogel I'd be happy to send you a copy of the procedure if you ever want to give it a try. Good luck with the synthesis. I can't wait to see it! Thanks for making these videos too. Maybe it will give me the motivation to retry the diphenhydramine synthesis someday; once I get around to finishing my rizatriptan synthesis some time in the next decade anyway.
You should stir your crystallization liquor with a stir bar at small scales. It promotes consistent particle sizes, mitigates how much you need to scratch the flask, and reduces impurity inclusion. So few chemtubers agitate their crystallizations. The real pro-gamer move is find the supersaturation window and pitch seed crystals then doing a controlled cooldown and/or antisolvent addition.
Watch glass is used for that to get seeding crystals for process initiation. Seedin is done on the flask wall right on the air-liquid interface. It does not normally result in disolving of the seeding crystals introduced.
Aromatic amines like to make purple oxidation compounds. Your dimethylaniline got oxidized during the processing. Reminds me of Perkin discovering mauveine (the first synthetic dye) by oxidizing a mix of aniline and toluidines with dichromate.
Considering the shelf-life of many of the painkillers commonly used for surgeries, having local processes in place to turn out pharmaceutical grade drugs might be prudent in case anyone decided to shut down trans-pacific shipping.
Tin reduction normally affords amino complex with tin chloride salt. Tin 2 chloride is used when you need selective reduction which is not the case for this particular nitro-derivative, most likely Sn metal cold be used instead of SnCl2 for economic purpose.
To get even more precise with a syringe you could try it like I do (note I'm a total armature with zero formal training.. so yea.. correctness/details on you). You just start with more air in the tube.. like I get that emptying it before drawing in stuff is, well, common sense.. but if you then you can draw in exactly 1.75ml (view un-obstructed by plunger), and then (assuming you keep it upright the whole time), you can flush it all out with the stored air.
Helional to MDP2P or piperonal. That'd make a great video. I know the MDP2P might seem suspicious but it's technically completely legal to possess. As long as there is no ill intent of course. I can help you with this. Id bet that would be one of your most popular videos.
I was very surprised at the idea that MDP2P might not be illegal (according to the DEA at least), so I looked at a bunch of official DEA lists of controlled substances and none of them mention MDP2P, PMK, 3,4-methylenedioxyphenylpropan-2-one, etc. However, almost all of these lists also say that while a given chemical may not be specifically listed, analogs and precursors are still illegal. I definitely would like to see how this would be prosecuted and how a defense for someone, say, producing and selling MDP2P would work in court. It seems like a gray area lol (which is why chemistry is awesome!)

Laboratory purification are realized by distillation in Kugelrohr.
