No, This method uses blending modes and there is no actual transparency right here, you'll always need any background to adjust your object transparency based on it! but you can save your object as PSD for any later use.
I got confused when you brought up the color and fill layer 4:12 as you didn’t show how as you did with the shadow and highlights layer. Did you duplicate the selection and then converted into smart object then added the fill or color layer to it?

Also what type of fill layer did you choose? I don’t why you skipped this stepped to create the fill and color layers
It's much better to add smart levels on the layers themselves (ctrl + L). No need to clip stuff.
Sometimes you need to mask levels and curves but with smart levels you have only 1 mask for all
