I'm a 4th year pharmacy student and want to be involved with pharmaceutical science research. I've been thinking about taking a Masters in Pharmacy, but I'm second guessing because from what I see the type of research produced is usually assays of plant extracts which I kinda don't like. By “fortify” do you mean purify? You need to do chromatic profiling, separation and purification techniques for that.

1 and 2. You can only use water in decoction method. You’ll be extracting flavonoids and phenolic compounds that are soluble in water only.
What is the shelf life of the "extracts" when performing extraction process? In my case, I consider Iyphilization/freeze drying the plant to achieve the same texture of green tea before boiling it. I wanted to store a large amount of dried leaves for future purposes and save it for decoction. Will it affect the shelf life?

Hey really nice video, so i wanted to extract anthocyanins from Jamun fruit using maceration with ethanol, do I need to dry the fruit or can I just simply put the fruit inside the solvent ?
Preferably dry the fruit to saturate the concentration of anthocyanin. Since anthocyanins are light sensitive make sure to dry it on a dark areas.

Hello! Thank you for your comment. To answer your questions, 1st. the reason behind freeze drying is to remove the water content from your extract without deteriorating your bioactive compounds. 2nd. if you want to obtain a liquid form of your extract, it's either you can dissolve the powder in your treatment vehicle (solvent used treatment, depending on your experiment), or you can use the extract automatically after decoction and filtration, but you have to make it more concentrated. If you don't concentrate your extract, it will probably yield lesser bioactivity.

There is no recommended ratio of solid to solvent when performing maceration because the end step is totally evaporating the solvent. So, as long as you have completely submerged your plant sample in your solvent then it’s good.

There’s no need to follow any ratio. As long as you have totally submerged your plant sample in your solvent then you’re good to go. After all, will remove the solvent when you run your mixture in a rotary evaporator.

@@kaeyaalberich4427 you just have to weigh certain amount of your extract and dissolve it in a certain volume of solvent medium.
Example:
10mg of plant extract dissolve in 10mL water will make 1 mg/mL plant extract.

Water is more polar than ethanol. If you want to extract bioactive compounds specifically those with antimicrobial properties, better check the literature of what Antimicrobial compounds present in your plant and identify in which solvent are they soluble.

Actually, mahirap ma-estimate kung gaano kadaming extract ang makakuha mo after extraction. What we do, usually, is prepare as much as we can.. mas maganda na yung sobra kesa kulang. Also, when it comes to the extraction method na gagamitin, nakadepende dapat sa target compounds or family of compounds na gusto mong pag-aral. As I said sa video, different extraction methods will give you different products... and these include volume of tincture/oil/concentrate or yung raw form nung na-extract mo. 😊
Hello! I am currently doing my research in plants. I have extracted it with ethanol extract and already airdried it. However, it contains chlorophyll and I am trying to remove it using a separating funnel with non-polar solvents. But there is a lot of confusion, can you explain how can I remove chlorophyll without destroying my active constituents in ethanolic extract also which non polar solvet is best or if there is any other methods.

I have a question, the powder that i have used for the maceration can i still reuse it? After filtering my solution, there was still leftover solute. After some time, it became dry again. And so i was hoping if i could use it again to make another batch of the maceration process. My plant material is very limited, you see. I used methanol as my solvent btw
You can but the amount of bioactive compounds might be lesser than the first batch.

thanks. one query is if you have prepared ethanol extract then how to incorporate it in feed during feed formulation.?
Before introducing you treatment (extract) to animal models you should make sure that all of the extraction solvent (ethanol) has completely evaporated. Usually, you can administer your treatment using the common drug delivery methods like oral, intravenous, intraperitoneal, etc. You can also add your treatment to the feed formulation but it will depend on what kind of feeds you have and its nature (dry/wet). Moreover, adding your treatment to the feed formulation might lead to inaccuracies in the volume or concentration of the drug (extract). So, you have to take note of that too. Like for example, if you want to treat your animal model with 200uL of 10 mg/kg extract, you plan how you can administer this accurately to all of your replicates to avoid biases and such.

Hello po! Thank you po for the informative video! It really helped us understand the methodology needed to conduct our research. May we ask po how much plant material (in grams) do we need to use to extract 5mg, 16.25mg, 35mg of the pure extract? And also how much solvent is required for maceration?
We are currently struggling to compute the amount of grams of plant material to get the according milligrams (We tried using the vague references in the video but still struggled). Also po we need to compute the amount of solvent used for maceration and the ratio is 1:2 (w/v). The volume of our final solution is 50ml and we need to dilute it for 3 concentrations (10%, 32.5%, 70%). After solving this, the grams needed for the dilution is already stated above (5mg, 16.25mg, 35mg).
To conclude, our group wants to know how much grams of plant material is needed for the final product using the 1:2 ratio for maceration and how much solvent is required. **We know that this request is a bit hard, if it cannot be answered, do you have any suggestions po on how we can compute it?** Thanks po doc!!

Yes, you can. But first, identify your target anticancer compounds. Next, determine in which solvent they are soluble. Then use that solvent to extract your compounds.

Essential oils are oils. After distillation, you will have the volatile oil of your plant. The term “essential” in essential oils refers to the fragrance of the plant that you’re using to extract the oil.

Around 1-3 days.. it depends.. you’ll know it’s ready if you can’t smell the solvent from your extract and you can observe that it has dried up.

I’m not sure but I think you need to perform steam distillation to get extract d-limonene.. try checking then form of D-limonene and look what steam distillation is. 😊
