No, TAS starts counting at the very first frame even if there's no input, and it finishes on the last input, which is the jump you do to get the star (not when bowser finishes talking). And your statement would be wrong anyway too since there's already inputs before the intro. ;)
In japanese version there are some stars shorter (Blast to the stone pillar, stand on the four pillars), short enough to probably save what loading would waste (around 6-7 secs at max)
So basically the reasoning of speedrunners playing the japanese version because it's faster is not true (at least for 0-star) and the difference between the two is, at most, about 30 frames.

In a 120 star TAS, lag difference wouldn't be more than 7 secs, and japanese version has some stars faster than American version, like Stand tall in the 4 pillars, or Blast To The Stone Pillar, so I'm still not sure which one would be faster.
