"Did they force them to read all of One Piece for these roles?" As far as I know, no, they did not. However, the cast for the main crew is definitely into One Piece properly. Luffy's actor watched a decent bit to get a good feeling of the character for his audition, and after he got the role definitely watched the entire anime. Zoro's actor had been following it for years (keep in mind, he's Japanese, and One Piece is just so big in Japan there's no way you don't know what it is if you live there) Nami's actress has been a huge anime fan for years with Nami being pretty much her favorite character ever and is fully caught up on the manga as far as I know. Usopp's actor I don't specifically know but it's clear he has at least some decent knowledge about the anime. Sanji's actor took cooking and taekwandoo classes to better get in the role and also definitely has seen a solid amount of the anime for it from what I know
Sanji probably the one who have the least knowledge on one piece, Sanji and Luffy both are new fans, but inaki caught up fast.. Zoro is japanese and Nami is long term weeb and maybe Ussop too but they definitely heard alot of rumors too

It’s also the fact in Japan spoilers are not classed as a big deal like they are in the west so media tours on Japanese media usually includes a lot of spoilers, just look at G5 being plastered all across train stations in Japan during the lead up to its reveal
they obviously know the entirety of the series/manga/anime because they have to read it and watch it to memorize and get to know the characters they are playing duh
No role requires that EVER. If an actor has learned everything about the role they are playing, they either did so in their own time or already knew the character prior to playing the role. The VAST majority of actors have no obligation to do any such thing, they just need to learn from and imitate the script they are given. They are absolutely NOT required to read or watch the entire thing just for some context because that is what the script is for.
All the mugis do they homework before they where cast for their roles, and i´m not 100% sure about Taz and Jacob, but Iñaki, Makenryu and Emily are real fans of the manga from a while.... the whole crew knows their OP xD
