Well, I dont think that it's an allergy. Because the medicine wasnt made of grapes, just grape flavoured. Someone who is allergic to banana's can still have items with banana flavour because artificial flavours are different from anything that is actually inside the fruit. For example: Cherry and banana flavour are one of the easier recreatable flavours, but the artificial flavour is smaller in size, a different type of molecule all together and is fairly inert to the human digestive system. Very few people are allergic to artificial flavours(in general, it's even rarer to be allergic to specific tastes), and of those who are, there are even fewer people who are both allergic to the artificial flavour and the original food that the flavour comes from. Too rare to base an easter egg on. The reality is simply that Goku did take the medicine which is why he is still alive when the androids arrive but due to him stopping he still got it. In dbza at least, in regular dbz, it's kinda unexplained why his heart disease showed up later, although odds are pretty big that Toriyama was pushed by Toei to make a super saiyan Vegeta(with a big reveal ofcourse, for merchandising) and needed a reason for Goku to tap out, thus him still having the heart attack.
To quote the Canon Goku... 'It comes from a need, not a desire'! True story!! Goku says this to Gohan in the HTC in Canon (spoilers to those who haven't gotten that far yet!! (should it really count...)) Vegeta, after realizing that he would never match Goku normally, gave up on surpassing him, leaving nothing but the need to get stronger, no matter what. That 'Pure' need overrode any desire he ever had until then, thus transforming him.
What's more is he would have died as he was in that asteroid field. His training pushed himself to his absolute limit. Faced with certain death, he stopped caring about becoming stronger than Goku and focused on simply surviving.
I bet anything that if Vegeta and Frieza faced each other at this point, it would work out exactly like it did when Vegeta had him on the ropes as a God. He'd have gotten desperate and would blow up the planet just to win. Cowardly bastard.
The way he described it, Vegeta gave up, then he became a super Saiyan. The idea of power coming from a moment of catharsis fits with the Saiyans trying to boost their power by controlling it better in the Cell saga too.

He had gotten rid of all the smaller ones but because of his training in 500 times gravity he was exhausted and couldn't destroy the huge massive asteroid that was coming. In that moment he stopped baring about trying to be better than Goku, and pretty much everything; even his own life. When he had finally and truly let go, his transformation happened.

I dont really think that's how purity works in dragon ball. It was his moment of full redemption though, as the rest of the show he was more of an anti-hero. It's kind of part of the show that Goku, Gohan and Goten are pure of heart due to being raised in relative innocence. Only using violence when needed or when being in a mutual fight, never really holding a grudge and always trying to seek the good in others. Vegeta doesnt show those traits untill Dragon Ball Super and even there he still lets his pride get in the way of his judgement. He acknowledges that his search for power should no longer be in comparison to Goku, because he knows that Goku is too dedicated to becoming stronger to the point where he doesnt really care about his personal bonds with other people. I mean, Goku never spends quality time with family to train, he never gets back in touch with friends unless they are strong enough to spar with, he doesnt even care about old teachers anymore as soon as someone stronger gets in the picture. Goku is kinda like a monk. No earthly attachments. He cares, but he simply doesnt feel responsibility.
The funniest thing about this episode is that people are chewing out Yamcha for not telling about the energy draining, but Krillin and Gohan also know. x3

It isn't necessarily a multiplier, I think it's a big addition to their base forms power. Even if Broly isn't canon, it wouldn't make any sense if a SSJ boost was a multiplication ^^

But after he was fully healed he got a zenkai (again) and was slightly above vegeta (again). then there was the hyperbolic time chamber crap, vegeta went in first, came out a LOT stronger than goku, goku went in, had a MASSIVLY more efficient training with gohan and almost went ssj2. Vegeta even went in a second time and could not fully close the gap.
