Ali Zafar was born on 18 May 1980 in Lahore, Pakistan. His father, Mohammad Zafarullah and his mother, Kanwal Ameen, were professors at the University of Punjab, Lahore, Pakistan. He has two brothers named Zain and Danyal.
Ali got married to Ayesha Fazli on 28 July 2009. He has a son named Azaan Zafar and a daughter named Alyza Zafar.
