I purchased your adapter and I now have my acrylic sheet with the 4” hole. Do you have any recommendations for the glue to secure the adapter to the acrylic sheet?
There's glue out there specifically for acrylic that dries clear, but a good quality super glue will do the trick. Sometimes it's dries filmy white though so you might want to do a test spot first or just be careful if looks are important to you. But it will hold just fine.
Where do you have your inline fan for your personal Glowforge setup? Do you have it hanging somewhere or sitting on a work surface?
I have a deep table and it sits behind the Glowforge.

I don't but may consider. Is 10" ducting pretty rare? I see a lot of 4" and 6".
Not for larger tents. You can get exhaust kits in 8in 10in and 12in.

I cut the hole at exactly 4"

Yes, I would go to your hardware store and get some window weather stripping with an adhesive backing and put it on all four corners.

Would this product work for replacing a glass pane in a basement window?
I don't see why it couldn't. I use 1/4" but you may need to use 1/8" because that's probably the thickness of the glass.

What would you do for an out swing door?
For a crank out window you’ll need a piece the size of your window. Replace the screen with a piece of plexiglass.

I don't sell the plexiglass insert just the plastic adapters that go in it.
