Aminoglycosides | Bacterial Targets, Mechanism of Action, Side Effects

  • Lesson on Aminoglycoside Antibiotics. Aminoglycoside antibiotics all have the suffix of -mycin, -micin or -cin and may be confused with the macrolides, which have the suffix -thromycin. Aminoglycosides are almost always used in conjunction with another antibiotic and are administered parenterally. Bacterial targets for aminoglycosides include gram negative coverage. There are specific side effects with use of aminoglycosides as well, which we also discuss in this lesson.

    Great video!! Simplified and clear.
    Kindly note that Aminoglycosides may be used for Aerobic Gram-positive bacteria such as most Staph aureus, Viridans group Streptococci and Enterococcus BUT they must be used in COMBINATION with another antibiotic.

    Great video, but the tricks are not accurate and can be confusing. You left out Vancomycin, which is not an aminoglycoside, but technically ends in "-mycin". Also, a lot of the Lincosamides end in "-mycin" and are clearly not aminoglycosides either. The other thing is Neomycin is technically an aminoglycoside and it is NOT parenteral. Otherwise, I love your simplification of a topic that can be very confusing.

    As a side note Aminoglycosides are not exclusively administered via IV. Especially, Cystic Fibrosis patients regularly inhale Aminoglycosides.

    I was injected the gentamicin directly into my vain back in 2004 and never forget about the side effect I felt right away. I never felt such a pain from an injection in my life. My arm was on fire, it felt like I was injected with peri peri or something like that. The pain was so intense that why I never forgot about the name of this injection.
    P.S. I was hospitalized 10 days for severe malaria and typhoid and was getting IV treatments.

    Sir how imipenem and gentamicin combination is used for pseudomonas infection .
    Both causes nephrotoxicity

    Wait,, according to this video aminoglycosides kills only gram negative bacteria ,,,but is is used in tuberculosis which is caused by gram positive bacteria,,can you explain this?

    According to some other sources aminoglycosides bind to 50s domain. Aminoglycosides bind to magnesium in rRNA on the 50s domain. What can you say about that? Have you heard about it? It’s strange that different sources give different info.

      Only streptomycin.binds to.30S. others bind to.either 30S/50S interface of 50S

    why aminoglycoside work a best in slightly alkaline condition ??

      Aminoglycosides, being weak bases, have optimal potency in alkaline media. The lower the pH the weaker the effect. If you want to exploit this, use a urinary alkaliner when using AMG's to treat UTI's.

    Why are aminoglycosides rapidly bacteriocidal but tetracyclines only bacteriostatic even though they both bind and Inhibit the 30s subunit?

      Aminoglycosides causes incorporation of incorrect aminoacids in the growing peptide chain..this will alter protein synthesis and alter cell wall permeability. Causing cell.wall.lysis and hence cidal

      Aminoglycoside are transported well in gram negative bacteria, they are given with pencillins to cover gram positive bacteria
