She doesn't really acknowledge Izayoi as an equal and sees her husband's desire to protect humans as a curious obsession she'll never understand, such as an average wife's passing acceptance of her husband's hobbies.
Sesshomaru’s mom may have hatred or disgust toward humans and half demons but her reaction to when Zero explained that if she dies, Rin(Sesshomaru’s wife) dies too. Proves that even tho she didn’t change her thoughts about humans and half demons… Rin has proven to be exceptional. (That’s just my opinion of course based on Sesshomaru’s mom’s sudden slight change of reaction from 1:15 - 1:27)
I think it’s also she has seen in the past how much her son cares about Rin. She cares about her son so the fact Lady Zero is willingly causing him pain brought up some emotion too. Think that’s why we got any eye movement AND frown from Sesshomaru’s mom.
It's more empathy and kindness for how short their lives are and how hard than in difference It's beneath her to look down on those who are beneath her
I think she is bothered that her son might be upset. As far as I've seen, she only cares about sesshomaru. So if rin dying upsets him, she will be upset as well because she doesnt want her son to be sad.

I can’t help but wonder whether the life-or-death link that Zero placed on Rin is similar to what Naraku did when he killed the priestess Hitomiko and imprisoned her soul using a demon spider-parasite in Ep 16 of “Inuyasha: The Final Act.” In that episode, Kagome had to learn to use her own hidden power through the sacred arrow to destroy the parasite/attack Naraku without harming Hitomiko. That same technique may be key to saving Rin from Zero’s spell. AND if there’s a second season (which seems likely), it may be Moroha who has to master that technique by learning it from her mother.
Or the new sword totosai has to make could have the ability to sever the connection which could belong to setsuna (or towa since it shows her holding it in the season 2 poster) because sesshomaru did ask why she didn't use her own blade and setsuna still has to get revenge on zero

So I think sesshomaru is sucking it up, made a deal with kirinmaru/zero to keep Rin asleep in a tree until he figures out how to save her. We will probably have more answers in the second season.
