Why do we add nitric acid

Is it 3-2 ml of HNO3(voice over) or one ml of HNO3(subtitles) ?

1 mL is enough ... and it helps when you have to neutralize the excess with concentrated NaOH solution to see the orange color.
2-3 mL is also fine, you get a deeper yellow color. As it is a qualitative test the amounts stated are only approximated.

if we added ammonia instead of NaOH does the color reaction still the same? Is it still positive?

Not sure about ammonia but ammonium hydroxide would give the same result

U can conduct it as follows
Add 1 ml of diluted egg albumin
      Add hno3 1 ml
      Now we will observe coagulation
      Then perform heating
