This is exactly the video I needed! I’ve just started a forensic microscopy lab at my university and I definitely want to have a concrete understanding of how to do this properly. I really thought it was something complicated but in the end it is a very simple set up I have been taught years ago. The problem was that nobody has ever told me the name of the procedure.

Why is there a tradeoff between resolution and contrast?
Hi Jaywai, thank you for your comment. When closing the field diaphragm or aperture diaphragm, we obtain more depth of field with more contrast but lower resolution. Have an excellent day!

Hi Jaywai! There is no particular reason related to this case, it is just a technical matter based on the characteristics of optics and light refraction.
@@semcify Because closing the aperture diaphragm also reduces the amount of light photons hitting the specimen and therefore the resolve isn’t so good. Simply less light in sacrifice for contrast and wider DOF. Good for finding invisible non-stained microbes
