It's simple dear. The liquid is physiological saline. Take 2-3 colonies of bacteria or amount according to size of colonies and volume of physiological saline. Mix it properly and read through this McFarland densitometer... If reading is greater than O.5 add extra physiological saline or less add extra colony...

If i don't have densitometer, can i use UV (i'll shake it vigrously before measuring) or not?
