Abuela removing the one white flower from Isabela's head is such an interesting hint as to how overbearing she is about Isa being perfect at ALL times. It really makes you sympathize with Isa, and understand the reasoning for her surface attitude towards Mirabel.

She hated Mirabel because she was jealous that Mirabel had freedom and she was subjected too more pressure. It wasn’t because of she was blind or anything.
I used to hate Isabela because reading everyone’s comments and analyzing her character made me see the movie but in a different perspective.
Also how forced Isa’s smile is. Early hint that she isn’t really on board with marrying Mariano.
And it was hilarious when Isa threw the flowers into Camilo's face

There's a theory that house may have the grandfather's spirit in it which would make sense with some of the actions like smacking Camilo like a grandpa playfully cuffing his grandson on the ear like stopping acting like a goofball.

I think that’s also meant to showcase the Isabella doesn’t want to marry him “especially since her grandmother is kinda forcing it upon her” and is meant to be foreshadowing to her hiding her true feelings. Especially with what we learn in “What else can I do” with the lyric “so much hides behind my smile” as we see after Isabella realizes what she’s getting into she tries to hide her fear with a smile but we still see uneasiness on her face.
If you also look closely, Isabela starts to breathe harder when Abuela Alma comes behind her. Either that’s her anxiety, she’s secretly scared of upsetting Alma, or both

The animation is just amazing in this movie. The way they animated the facial expressions is awesome and so well done.

I find it sad how Alma seems to value Isabella having lots of children to carry on the miracle rather than her happiness and well-being in her forthcoming relationship with Mariano.
There is a UA-cam short which mentions that Isabela looks a lot like Alma as a young woman, and Mariano looks like her late husband, which would theorize that Alma wants to live her ideal life through Isabela.
It's also quite valued traditionally in Spanish culture to have quite large families. Though, this is changing in modern times.

I notice when they’re talking about the announcement Isabela really starts to breathe much heavier than usual. You can tell she don’t wanna marry him.
I like how Isabela represents someone most siblings would be jealous of - beautiful, powerful, loved and envied by those around her. But in truth she is unhappy. Some even theorize that she is jealous of Maribel for not having so much pressure set on her, and for having more freedom.

I feel like Dolores claimed that Mariano wanted five babies just to make Isabela feel scared and so she can be subjected to additional pressure. The white flower indicates that Isabela doesn’t actually wanna marry Mariano and decides to make a smile to make Abuela and the family happy.
Guys in 1:39, as you can see, pink flowers appears on isebela's head. But there is an Odd flower too, and the odd one is the white flower. The real isebela is not that pink flowers. Real isebela is that white flower. And abuela sees that there is an odd flower on isebela's head, and takes the odd flower from her head. Because abuela wants a perfect Isabela.

Fact: Not only Mirabel mistook Camilo for Dolores (because of his powers), Mirabel unintentionally let him know about the problem with the magic. 😶
But he doesn't seems to care about it-

Fact: Mirabel says out of all my older cousins your my favourite cousin to Dolores. Dolores is the only cousin older than Mirabel since Luisa, Isabella are her sisters, Antonio is younger and Camillo is the same age.

You know something I don’t get? The madrigals serve their town even though it was abuela’s candle that saved them all. You’d think things would be in reverse. That the community would serve them instead. Consistently tried to prove that they deserved being saved in the first place

The community is very dependent on them which is why the fuss later on when the magic is in trouble. And why they thank the Madrigals for generations of aid by helping them rebuild. The magic is with the Madrigals so they are the head family of the town. Also Colombians are Christians, traditionally Catholic, it would be blasphemous to worship them and serve them as gods. There’s literally a town priest.
It's called Survivor's guilt. Alma/Abuela blames herself for Pedro's death (because he sacrificed his life so that she could live and raise their children), so, in her attempt to make amends, Alma has convinced herself that her family was granted the Miracle, (the Encanto, Casita and the Gifts) to serve the village and prove themselves worthy of Pedro's sacrifice. It's not her fault, she never had the time to grieve properly, as she had to raise three small children, and the rest of the village was as traumatised as her and elected her as their leader due to them feeling protected by the Miracle. No one was there to tell Alma that she had got a Miracle because of Pedro's love for her and her own love and ultimate attempt to shield the triplets with her own body (fully aware that she and the babies would have been killed next), nor that her triplets gained Gifts due to the FIVE YEARS OF DEEP UNCONDITIONAL LOVE Alma gave them (hence why every Madrigal gets a magical Gift at the age of FIVE).
This is party the cause of Alma alternating cold anger and pity whenever she interacts with Mirabel: she is reminded of a time when she was non-magical and WEAK, when she lost Pedro. And Alma had been fighting for years to be strong. In her opinion, if Mirabel didn't get a Gift, that meant that her family at the time was no longer worthy of Pedro's sacrifice, that was WEAK and Alma was in danger of losing everything. Mirabel represents that weakness: she's Giftless and seen as lees than the others.
The other reason for her harsh treatment is that Alma BLAMES Mirabel for Bruno disappearing: the night he went away, Abuela had asked him to look into Mirabel's future to know what it meant for the magic, then went into his room to see the prophecy and found Bruno missing. She didn't know the content of the prophecy, but she knew that Bruno had made it and that involved Mirabel. The anger she shows Mirabel is genuine, while the one she uses while referring to Bruno is a mask: Alma states "Bruno didn't care about his family!" (actually she meant: "Bruno didn't care about his MOTHER!"), but, when Antonio keeps her seat warm, Abuela calls him "Tonito" because she's reminded of how much sweet and loving Bruno used to be with her (and she used to call him "Brunito").
By the way, the Madrigals are having breakfast in the garden and the food table is outside, so... Is Bruno outside too, maybe hidden in the bushes behind Pepa and Luisa? Maybe he woke up early and sneaked outside, then as Julieta was setting the food table, Bruno stole some arepas and bananas, and hid back in the bushes! It's enough near the door to sneak back inside later and enough near the family to listen to their conversations! Plus he wears a green ruana and can blend into the bushes! That would be both funny and deeply sad: Alma walks past her son without seeing him, and, after she says "Tonito", she looks around for her Brunito... who is actually there, BUT UNABLE to come out to her.
