Thank you. Please how can determine amount of silica gel to column if you have example 0.8 gm from mixture? This if you synthesized compunds in lab and need to do column chromatography. Tq

There are a lot of variables involved that I do not know. How complicated is the separation? This affects how long the column needs to be. The amount of material separated is related to the diameter of the column. So there are at least these two factors involved. Without trying it, I cannot say.

Can we use CaO in place of Alumina?

You could use silica gel, but I wouldn’t recommend CaO due to its water reactivity.
