In that story, there is a masquarede with different colored rooms, with each room representing different personality. So this line is referring to how Joker can switch Personas to basically change between personalities.
At his unmasking, the Red Death, a bony and wasted figure speckled with blood, had no real form beneath his disguise. The Red Death swept through the masquerade, killing all attendees and the host. Sometimes, you don't want to look underneath the mask.
I believe the song is actually about Akechi/in his perspective, since hes the one who's actually putting on a mask to the world and hiding his true identity, his persona is also a shape shifter lol

I remember performing this exact song in my singing class, my class was absolutely captivated by the song, and by how i handled that performance, it was awesome getting all that attention, and what made it all the better was how solid that performance came out. I contemplated actually wearing a masquerade mask for this, but i opted against it, as it’d take away from my performance, but once i had finished it, my class said that I legitimately “took off a mask and bore everything out for all to see” craziest experience ever, and it changed the way i see myself because i don’t open up to people easy.
