Cloning Agarikon to save its Mycodiversity

- One way of protecting the biodiversity of fungi - mycodiversity- is to clone a wild specimen and get it into culture.
Over many years we have built the largest collection of Agarikon (Fomitopsis officinalis = Larificomes officinalis) strains in the world. Agarikon habitats are increasingly being lost as this mushroom dwell primarily in the old growth forests of northwest North America, in sky islands in the Alps of Austria-Slovenia-Italy and in northeastern Eurasia. As the forests are cut, unique strains of fungi, including Agarikon, are lost. Agarikon is now considered a threatened species in Europe.
By preserving as many Agarikon strains as possible, we hope to test each one for their unique properties.
The first step is to find them. Pamela Kryskow, MD and I hiked deep into an old growth forest near a lake on a remote island in British Columbia. Once we found a colony, we used a cork borer to excise a small piece of tissue and left the conks to live.
The next step was to go into the laboratory and subculture fragments of this tissue into nutrient filled petri dishes. PTC
Then, after cultures grew out, one or two are selected for further culturing into another generation of dishes.
From one of these dishes, the best-looking mycelium was then transferred into test tubes for long time storage.
With that last step, we have "banked" the strain once it grows out and then put into refrigeration for long term cold storage.
In these four videos, we show you how we do it.
For teachers, students, remote learners and the DIY people out there, I hope you find this useful.
cheers,
Paul Stamets & Pamela Kryskow, MD
on Remote Island in British Columbia
People in the future will realize how important this man has been
@N B the strain of mushroom or man? If mushroom agarikon has a strange history, fairly newly discovered, I. Fact Paul discovered and named his own species
I should say newly rediscovered. Very fascinating species to do some research on! Lions mane is another fascinating species. Theain reason for it's scarcity is it needs old growth forests to thrive, and we the people are busy cutting all those down. I had a dream last night that the whole world was a ant colony, that knowingly destroyed their habitat
Thank you Paul .🍄 Please make more videos

For sure, I love this man. He should be spoken about alongside all great names, Einstein, Darwin, Hawkins etc.

Why have you not be awarded the Nobel Prize? Thank you, thank you, thank you.
He’s not famous? He’s not saving people’s money?
Because he has dishonest practices with his brand. He’s not a mycologist, he’s a salesman and a very good one at that.
@@DennyMapleSyrup Hmmm, are you a hater, jealous, moron, or all the above?
@@michaelcorbin oh don’t get me wrong, great marketer, I’m just a huge supporter of the actual culinary and medicinal mushroom community, which he is not part of. He’s a snake oil salesman, join some culinary mushroom groups on any social media and ask about Stamets, you’ll see what I mean. I think a better question would be are you blindly following Stamets?
@@DennyMapleSyrup Hey Dave... Have you heard about his discoveries linking the shortened lifespan of bees to the lack of available mycelium do to rapid deforestation of the old growth forest? How about his solutions to this problem? What about his discovery involving cordyceps spores as a completely safe alternative to chemical insecticides?
Or his discoveries involving myco-remediation, cleaning up oil spills, voc’s etc. You know..... just restoring the ecosystem and saving our planet. But I think you should listen to the people in your Facebook group.

Great work Paul!
I recently revived a few cultures which you prepared 15 years ago in similar master slants and I can verify 3 out of 5 have been successfully revived after sitting dormant in a fridge for over a decade!
Salute ❤
The entire planet should rally around this man 🌎 the potential from his research should be realized and the information needs to be common knowledge asap! ❤
our future generations will make this go viral with appreciation
Thanks for what you do, Paul! I've harvested mushrooms for food and medicine for years in Ohio, but you opened my eyes to a whole new fungi world! Just received your book Mycelium Running, and it's amazing. I work with troubled youth, and we have a pond on the property. Im gonna try and use some of your methods to teach these kids a whole new way of taking care of our planet! Most are just smart kids being dumb. A product of their environment they learned in. Usually horrible parents, and kids having kids.
My great-grandmother was full blooded Cherokee. Thank you for talking about the 7th generation. We need to talk about that way of life more and more today! I just had my first flush of lions mane. It was my first attempt growing any fungi indoors. Success!
Thank you Sir!
Sincerely
Rodger from Ohio
I hope Paul has enough colleagues and apprentices that can also help carry this torch! Inspirational
If he doesnt im one call away if i could ever be so lucky. Truly inspirational
Hopefully next year i will get my mycology degree in Brazil and Paul inspires and shares much knowledge
I completely agree with you! I’m learning as much as I can right now and want to keep going. We need to grow this knowledge. It’s so important
We just found our first lions mane in the wild. So much fun hunting and photographing fungi. Thanks for all you do!!
I feel such a privilege watching Paul Stamets do this himself just 2 weeks ago. People say 2020 was bad; I say it was great because we know more about preserving our own species AND I stumbled upon Paul's work this year.
FYI: My 1st agar attempt is sitting on the shelf behind me. wish me luck!
Wow, this is amazing! Thank you for sharing this with us all! I am born and raised in Port Angeles, and I remember seeing some when hiking in some of the old growth ❤ 😊 so amazing to know I have seen them! Thank you for keeping this knowledge alive!
They're alive and well in the Marblemount area!
I saw two batches, pretty big colonies on the Hidden Lake trail if anyone wants to go take some pictures!
Paul, this video's content is perfect and rich in quality advice and techniques. I'd love to see your tips and tricks on initiating fruiting
I have a bacti incinerator for my scalpels now because of your workflow in this video!
Science!
I can't think of a single living man for whom I have more respect than I do for you Mr Stamets. I hope to meet you one day.
I'm an aspiring biomedical research student and you have driven my inspiration for mycology in the context of food science, I love to learn from you. thanks for sharing.
I love how much care Paul takes. Not to mention how he makes backup of the backups of mycelium.
redundancy is important in any robust backup system, I appreciate seeing it
Especially with Fungi.
You are changing the game Paul. You’re doing the “lords” work.
@ɮᴇᴄφᴍɪnɢ - WR if you've nothing good to say shut the fuck up
@ɮᴇᴄφᴍɪnɢ - WR Mr. Stamets is educating, spreading love, and doing what he can to protect life on this planet. That may not be anything new but it's extremely rare and more admirable than most could ever even aspire to. I implore you to take a hard look at yourself and try to gain a perspect of love instead of one of toxicity and hate. We need more positivity in this world right now and a lot less trolling.
@ɮᴇᴄφᴍɪnɢ - WR Come man, no need to kill the mans message, he is being nice, maybe you should learn to be too, nastarovia God bless.
I agree.
@ɮᴇᴄφᴍɪnɢ - WR what ever you give it will turn back to you multiplied fucking moron. you said one negative comment and you got already times 3 back. I have no doubt this happens in your life generally. What did you do and had good impact on our planet today?
Awesome stuff Paul! Gonna attempt to capture as many different mushroom genetics in Colorado as possible this year thanks for the knowledge.
TYVM for keeping our little slice of paradise just that much more special. Makes me want to look in the forest of my backyard on the same remote island!
So great to have a peek at how Paul works. Would love to have a flow hood like that!
Wow 80 phenotypes, time to start hybridizing for that super strain!
Hybrids are great. However the highest works is to preserve original genetics and innoculate the local areas with original indigenous genetics.
Now that is a flow hood I am envious of. Great video Paul and great work as always
We have an old fir stump with some type of these beautiful agarikon and had no idea how special they are! Thank you for the magnificent work you do. It continues to inspire our curiosity and has lead us to discover these as well as many other mushroom varieties around our southern Oregon homestead.
Keep up the great work Paul! :) You're truly making a difference in the world, we all appreciate it!!!
Thanks paul. Im always keeping an eye out for them when im in the woods. I know some spots with huge ones near mt adams. I dont tell a soul. :)
Haha
Paul, you're going to save this world. It could be in a year or in a century, but you'll be the reason for a hope when all else has failed. Your work is incredible
You have installed a new interest or even passion in to my heart Mr Stamets. Much love from Scotland🙏🏻🍄
extremely wholesome vibes thank you Paul! Always so informative
Paul is so amazing, it's insane how important this is to us
Thank You for all you do while being kind and humble enough to teach others along the way!
I haven't seen one of those instrument sterilizing kiln things since the last time I was in a biotech lab. Really brings back memories -- not necessarily fond ones, but memories nonetheless.
I seen a very large one on my walks I definitely check on it every time I pass it
Thank u for everything you do 👏
I'm so jealous of your flow hood!
I tried to save a very particular (and suspected) strain of liberty caps before their habitat became industrial farmland. It was incredibly potent, maybe averaging on the double of the liberty cap potency overall. They could grow to very large size, the heaviest fruit specimen I picked weighed 60g wet and had a cap large as my hand. They were also constantly bruised. I tried to populate another field with them but they didn't propagate at all there. I miss that strain a lot, and I don't want to see incredible species of mushroom ever go extinct again; edible, poisonous or psychoactive, or otherwise.
Thank you for all that you do, Mr. Stamets. You're a true inspiration, and I wish you nothing but continued success!
Do appreciate all you are going! Keep researching!
Thank you for being Paul.
Paul is a genius in his field of work. One day hopefully i get to shake your hand sir. Thanks for doing what you do for the mycology science 🍄🍄
I love watching your videos Paul you have inspired me so many times.
Paul has shown me alpt during my Psilocybe Cubensis experience tonight and I love this guy, Father Nature
Amazing video....I recently was called by the Forest, off trail, and found 2 Trees with Agarikon on the same day (after looking for years since I heard about it from You). I was looking for a video like this. Thank You very much. One of the Agarikon Trees I was called to, had quite a large Ghost Agarikon Laying on a bed of decaying wood chips at its feet (holy wood chips from the long dead, decaying Tree with Agarikon). I sampled a tiny piece of the Ghost, as there were many small chunks strewn about the floor, as well as few larger chunks. I felt the swelling in my knees vanish almost immediately. A truly incredible anti-inflammatory....or an incredible Placebo I don't know. The Agarikon which is living, smells very nice and lemony. I noticed tiny white chunks of the Agarikon were scattered and buried around the Tree, and don't seem to be decaying at all (could be a truly potent anti-microbial). I must have run/walked passed this Agarikon, which was not far at all from the Trail, hundreds of times. And I found another one earlier. Truly a Magical Day. Anyhow, thank You for your Amazing Work, I look forward to more videos and more incredible information. I will share this. How do we go about getting you samples off these Holy Agarikons? Thank You. Cheers.

I see agarikon in nearly every location I have been, along Oregon Coast, as well as inland 70 miles.
Some places have several on one, or more trees.
Paul Stamets, I first heard on radio, about 15 yrs ago.
Brilliant work, he does in the mycologic world😎
That flow hood is epic
Thank you for sharing. I hope whole genome sequence of your library will be open sourced. That is what I gonna do once I have Nanopore MinION figured out. MushLove!
Agarikon has been the only mushroom supplement I've taken that I've had a reaction to. Gave me shiners blotchy skin every time.
Been harvesting this for a long time, it’s a medicine with no question.
