Bilirubin is a yellow pigment in everyone’s blood and stool. Sometimes the liver can’t process the bilirubin in the body. This can be due to an excess of bilirubin, an obstruction, or inflammation of the liver. Direct bilirubin is elevated in obstructive jaundice and cirrhosis of liver whereas indirect bilirubin is elevated in prehepatic disorders (hemolysis of impaired conjugation or transport) in the liver. Estimation of bilirubin can be done in serum/plasma using Jendrassik & Grof method.

Sir, you may have to refer the kit insert or IFU of the manufacturer whose kit you are using. The requirement of blank will be mentioned in the IFU by the manufacturer.
