    Another excellent and informative video, thank you Michael.
    My first thoughts were that, as with other issues such as NAD+, it would be great to find out WHY the levels fall with age so we can try to fix it rather than topping up a leaking tank. That said, in the meantime I'll happily top up that tank if it'll get me home! 😁

      It declines because of the rising CD38. CD38 requires ~100 molecules of NAD. This is the reason. Not a scientist, but if you boost your thymus and thus your immune system, you could lower cd38 and raise NAD.

    I would like to underline about something that is not mentioned in this very informative video.
    After 12w without GlyNAC supplementation, total RBC GSH levels went back slightly above baseline (0.9 vs. 0.8 of baseline). But inflammatory and DNA oxidative stress markers haven't reach near to base line high values. They were significantly better (yes, not near to "young" levels) but GSH value was only 0.1 better and not close to "young" levels.

    Excellent video, great information.
    I knew NAC was good for you but.. wow.. 9g per day? I've taken up to 3g per day and felt strange because of it. Creatine gives me a similar feeling when taking high doses of it (more than 5g).
    A sort of bloated / swollen feeling.
    I've been taking 1g NMN, 2g TMG & 1g Apeginin daily for 9 months now and that has been a game changer for me.
    Looks like I need to add NAC and Glycine to my daily routine.

    I have been taking glycine 3x per week since last Fall along with other basic molecule / vitamins / mineral supplements. I think the synergistic effect which I notice is the improvement in cognitive intensity and duration for heavy strength training.

    I have misgivings about glycine supplementation in the context of some neurodgenerative diseases (multiple system atrophy), especially big bolus doses. There was research by Lane in the early '90's on it; Lane's case study of an MSA patient with a glycine transporter defect was interesting.

    Hopefully the next study will find the minimum doses required to get these results. This would be very expensive to repeat at home.
    Also, why wasn't glycine (which dropped the most) given more emphasis/supplementation relative to cysteine?
    I would like to see a similar study with only glycine supplementation (TMG and no NAC).

    9g per day of NAC ??? ... that's a massive dose. I take it and feel odd if I get anywhere near 1g (a sort of brittle feeling, difficult to describe)
    WebMD doses for all reasons go nowhere near 9g. I'd like to see a study with a lower dose.

      In rat studies high doses of NAC reduced oxidative stress etc but gave the rats lung cancer. i dismissed it because the NAc amounts were so high in comparison, but if it takes such large NAC to produce positive results the risks might not be worth it. i hate all these unrealistic dose studies.

    They dosed the NAC at 100mg/kg which seems very high as 1800mg is the upper limit of recommended dosage. Is this to try and overpower the breakdown of NAC in the gut and short half-life? If you used, say, Liposomal NAC, could you lower or significantly lower the daily dosage? How would you safely test this on a N=1 study?

      The safest way is to start with a low dose, but also with full panel blood testing at each step. That's what I've done with NMN (see the NAD video series). Then, one can identify if it works, or is neutral or detrimental, even at higher doses.

    This was a very interesting subject! Your voice sounds strong and healthy and I imagine your diet has been protective during these times. I too, don't like to supplement, and although i may not be able to achieve the levels of glycine in the study, I can up my levels more than before! Thinking of buying bone broth. What are some other recommended foods to achieve at least higher levels than we had before. I think sprouts too are beneficial for upping glutathione. Could this be related to why older adults need more protein as their ability to absorb these amino acids might be reduced? Thank you for these encouraging videos. Each day we have the potential to be better than we were the day before! Forgot to ask about increasing NAC through diet...

      Thanks Pamela. As you mentioned, supplementation is a last option for me, but based on the doses used, it may be less likely to boost glutathione through diet alone. For ex., my normal intake of glycine+NAC is 4g/day (but not including the amount in sardines, which isn't listed in cronometer). Based on what they used in the papers, my intake is about half needed to maximize GSH, at least in older adults.

      @@conqueragingordietrying123 I have been supplementing with 1.5g/day of NAC for over a year so I know it's safe but have probably been misinformed about safe dosage which your video indicates to be about 9g/day. Also take 2g/day of glycine which I will increase to 4g/day. If you can confirm a recommended dose this would help. I am in excellent and improving heath at near age 70. I take full responsibility for all my own actions so please be assured that no legal accusations will ever be made by myself against you for any recommendations you may make to me.

      No worries,@@rustybolts8953. In terms of a recommended dose, that would be a guess based on the published literature. Beyond that, I'd recommend blood testing to determine if your current dose is detrimental, neutral, or beneficial. With that method, you'll need a few tests (assuming test-to-test variability), and then, altering the doses in conjunction with blood testing can determine how much may (or may not) be beneficial for you.
      For ex., see my approach with supplementation vs homocysteine, where I'm now only taking methyl B12 instead of methylfolate and B6, which are not significantly correlated in my data:

      @@conqueragingordietrying123 yeah, I can't afford a lot of supplements. I eat an avocado every day and about every other day, I wilt a huge cello container of organic spinach. I usually have 3 eggs for breakfast and sardines about once-2x a week. I make my own 10 hour bone broth...just toss all your chicken bones in a freezer bag and save up til u have a bunch. all good for glutathione levels. Fasting is THE wonder drug for anti-aging...saves so much money, I can often afford the organic, or grass fed options!

    I read the study some days ago. I was already a NAC "fan" and glycine too but I was perplexed by cysteine amount which was very high (around 7g for 75kg body weight as I recall), what do you think?
    BTW I have to lower alcohol consumption, it seems in time of COVID bad habits are easy to take 😉

      The NAC (and glycine) amounts that they used were indeed very high. Also, these findings were in 2 small studies, including a total of only 32 subjects.
      The same research group is performing these studies, so replicating the supplementation approach in other labs would be nice.
      Nonetheless, there are a lot of promising findings, many of which I showed in the video.

    Is sugar oxidation more damaging to the body than fat oxidation? E.g. Is there more ROS generated when sugars are oxidized relative to fats? I've heard athletes can't be as explosive without glucose or glycogen stores - typically more power dense fuel is less efficient fuel (more byproducts or heat).

      I'm not trying to demonize sugar oxidation for energy, which has its place, including high-intensity exercise. However, we spend most of our existence burning fat. That mitochondria in older adults have worse fat oxidation, and correspondingly, try to account for it by being better at oxidizing sugar doesn't fix the problem. Note that youth is characterized by mitochondria that are better at burning fat, not glucose, and restoring GSH improved that in older adults.

    I take these supplements. The dosages mentioned are multiples of the supplement directions. 600mg NAC would go from 1 capsule to 15.5. 960mg Glycine from 2 capsules to 7.3 capsules.

    Michael there is a study where chamomile increases glycine. Do we need to supplement glycine if we drink chamomile?

      Camomille contains Apigenin (but not as much as Parsley). It reduces CD38, which is a big consumer of NAD+.
      But I don't know if it does anything else. I doubt it does much for Glutathion, though.

    Btw, I thought the (anti-)oxidant theory of (anti-)aging is dead? Does this study & similar study on HIV+ & mice/c elegans studies suggest otherwise?

      Ha, I wouldn't say it's dead, but the origin of that theory was that it was/would be the end-all-be-all of aging theories. From that perspective, it's not, but it's definitely a contributing factor during the aging process. Nonetheless, there's lots of positive data on other aspects of aging that may be more important, including inflammation, insulin sensitivity that are positively affected by GSH restoration.
      Now, the major question is, will it work in larger populations, or is this an artifact of 2 small studies (n=32 total)?

    One of the key factors is dose. It's a fairly high dose to get these results. The studies show the dose in mmol/kg of body weight. I've converted mmol (milli-moles) to grams to make it easier to figure out the dose.
    As an example: I weigh 80kg and to get the results in the study I need;
    1) glycine - 0.10 gm * 80 kg = 8 gm - cost in bulk powder is about $0.18 per day
    2) NAC - 0.132 gm * 80 kg = 10.6 gm - cost in bulk powder is about $1.96 per day
    Dose matters, they (do you know this is a division of Nestle doing these trials) also did a trial with similar results with less but I'm more interested in maximizing my results
    If you are paying more, you need to shop around 🙂

    I'm surprised they supplemented so heavily with NAC, when the decline even by older ages is only 24%. It seems like glycine is the real problem, given it's nearly halved.
    I'm also surprised you don't supplement Michael. You're getting up there in age. ;)

      Thanks Aldous Orwell. Someone in the comments posted a paper showing that oral GSH raised blood GSH, so you may want to check that out. In terms of which approach is better, however, no studies have compared glycine+NAC vs direct GSH supplementation.

    Does TMG equate to Glycine? I take 3-4 grams of that daily. I did see one study that foundTMG increases Glutathione in Rats

    This is a very striking result. It's not clear from the video if the dosing used for NAC and Glycine was optimal; it seems possible smaller amounts would be adequate. Also, sulforaphane is supposed to encourage the enzymatic production of glutathione, and could also be beneficial as an accelerator for this.
    Collagen peptides, made from cowhide, have a large percentage of glycine. I get the grass-fed type, and upped my daily collagen peptide intake from 13 g to about 20 g, which includes about 5 g of glycine. I also started supplementing with NAC, 2.4 grams/day (1.2 grams twice a day).
    There is an argument that humans traditionally ate much larger amounts of glycine than they do now. This is because humans traditionally ate animal parts other than just the muscle, and these are much higher in glycine. For this reason, the body can only produce something like 3 g of glycine/day - most of the glycine needs traditionally came from the diet - and 3 grams isn't enough in the long term.

      I'm not familiar with any studies using it to impact glutathione, but it would act as a cysteine donor, which could increase GSH synthesis. However, that's the reductionist version, I'm not sure what the broader systemic impact would be.

Anyway, there's the following paper where they gave 2 and 4 g per day of g-GC: where it increased lymphocyte GSH by 53%: "Oral administration of γ-glutamylcysteine increases intracellular glutathione levels above homeostasis in a randomised human trial pilot study." by Zarka and Bridge. I shared the PM ID in another post, but UA-cam deleted it for some reason.

    glutathione also downgrades graphene oxide in vaccines, it increases the nutrophils in white blood cells and oxides the graphene oxide similar to how the natural hydrogen peroxide in cells work to granulate bad cells, foriegn bodys and viruses,
    im interested in the bonded glutathion you spoke about in the video, amino acids are key to good health and it is so complicated there are so many, the perfect thing is a full bredth whey proteine shake with added BCAA's branch chain amino acids, eat mediteranian diet and have a shake once a day, also will help the global food shortage.

    Is it possible to get these results taking SAGlutathione? Or a combination of lower doses of NAC, glycine & SAG?

      I'm not sure which approach will be best for optimizing blood glutathione (GSG), but GSH and GSSG are commercially available for measurement in blood. One can try different interventions in conjunction with blood testing to sort it out...

      Yeah, it's a very large amount. Basically 8 pills a day of each supplement. It might work (and be more affordable) as a shake if one used one of the large bulk powdered supplement vendors. This would require that both substances be relatively flavorless; NAC definitely is not flavorless (it's bad), not sure about the glycine.

      Ha, I'm not anti-supplementation, but I favor targeted supplementation when there is a need. Inflammation and insulin resistance increase during aging, whereas GSH declines, and if I can't reverse that with diet, exercise, etc, then I may take glycine+NAC!

      Glycine is involved in detoxification reactions. So more xenobiotics-->more glycine conjugation-->potentially less glycine available for GSH synthesis.

      Glycine is a major part of Collagen. So, that's very likely the reason why it's to low. We doesn't eat as much Collagen as we used to.
      The level of Glycine they supplemented with (7 -- 8 g) is about what we could get from eating the old way.
      Supplementing with Collagen and especially Glycine is a good idea!

    Thanks for the good video, should we be worried about supplementing NAC due to the study of Vitamin E and NAC increasing liklihood of lung cancer in smokers? It may have just been the Vitamin E which caused it but still a worry.

      Cancer risk with vitamin E (alpha tocopherol) supplementation was found to be dependent on COMT genotype in women. It lowered risk for those with met/met genotype, but raised it for val/val.

    Do we know what the typical Glutathione levels are of 40-50 year old subjects are? Perhaps the middle aged would need less of a raise of Glutathione to get to youthful levels and lower levels of supplementation.

      Thanks Viracocha. I'm not anti-supplements, but I favor target supplementation. For ex., I take L-thyroxine for hypothyroidism and Vitamin D in the winter. In terms of senolytics, based on the data in the video, the combination of glycine+NAC may be the best of the bunch, and if I have deficits in inflammation, insulin sensitivity, etc., this combination may be what I take.

      @@Avital4414 Hey Rt L. I'm not a fan of general recommendations, but individualized testing to find out the best approach for you. When considering that glycine+NAC can improve insulin sensitivity, one would expect lower fasting glucose, or lower post-prandial glucose as a result of supplementation. That may be an easy way to identify when would be the best time to take them.

    Judging by the insulin sensitivity and glucose slide, these older people were not metabolically health to begin with. I would like to see the study done with people who start out metabolically healthy. Not likely, but still, I would.

    So many hallmarks of aging altered!
    Do you think taking liposomal glutathione would be better or worse than the Glycine + NAC combination?
    What might be they cause of declining Glycine and Cysteine as the years pass?
    Some of the improvements seemed to remain over baseline figures when the supplementation was stopped. Might that indicate a marginal long term improvement has been achieved?

      Thanks Luca Crane. I hope the data isn't too good to be true, as there's only 1 research group using the high dose glycine-NAC approach. In terms of that vs. liposomal GSH, I'm not sure, no studies have compared which would be better in terms of increasing systemic levels of GSH.

      @@powerpuffherk This is speculation, but I'd bet on increased glycine conjugation, which serves as a detoxification mechanism. "Bad" metabolites in the blood increase during aging, which are then conjugated by the liver and kidney with glycine to target them for excretion. However, that limits the amount of glycine available for GSH synthesis.

      @@conqueragingordietrying123 I've been suffering and living with chemical sensitivity headaches for six or seven years now without being able to determine their root causes, or how to avoid them (other than constantly avoiding artificial odors in my day to day life). They trigger tension in the base of my rear neck, which leads to the headaches that can only be treated with hours laying down, or muscle relaxants (naproxen, ibuprofen, acetaminophen, nor aspirin make a dent in the pain). For reasons unrelated to these headaches I decided to incorporate liposomal glutathione to the three or four anti oxidants I already supplement with on a daily basis. To my surprise I realized about a week later that I had not had any headaches. I specifically realized it on an occasion where I had been exposed to an odor which had always caused me headaches in the past. I cannot wrap my head around the correlation between the glutathione and the dissipation of my headaches. I know that glutathione helps the body deal with free radicals and oxidation released by inflammation, but that doesn't help me figure it out. Can you connect these dots anywhere and see where the correlation may be? I feel that if I can figure out how these two things are related, I may be able to get to a deeper root cause. Thanks!

      Hey K J, unfortunately I don't have a recommendation-I'd like to think that all glycine and NAC should be of the same purity, but that may not be true.
      I don't (yet) take Glycine + NAC, but based on the human data (albeit in small studies), it may be the best option for anti-aging, until there is comparative data against rapamycin and/or acarbose.

    Dr. Lustgarten Very good information! My interests are in two areas: 1) Increasing glutathione, and 2) New research in Gut Health, which is also quite exciting lately. Some researchers are talking about returning missing strains to the gut that have some cool functions.
    Personally, I shall probably use Liposomal Glutathione (1500 mg) with NAC at 5-6 grams, and glycine at 12 grams, and I also take in cofactors like Natural vitamin C, selenium, and natural B vitamins from Beef Liver. Thinking about shortly adding yogurt batches with new strains, and I already eat several fermented veggies. These studies made a great touchstone though.

      Do not use Liposomal Glutathione as it can cross the blood brain barrier and cannot be metabolized. just use glycine and NAC and you should be able to make plenty of glutathione.

    Did they identify why the older population had less cysteine and glycine? Was it because their stomach acid is lower maybe and they don't break down protein as well into amino acids? Is it because of other micronutrient deficiencies that influence digestive enzyme production so they tend to eat less meat (e.g., zinc)?

      Most good protein sources give you perhaps 2 g of Glycine. That's not enough!
      Collagen has about 23% Glycine. So that's our best source of that.
      But you need most of it for building your own Collagen.
      Supplementing with Glycine is a go9d idea!

Clearly need to get good price for this . Glycine has multiple good effects and can take much more than 7 g for other issues. Sharry Edwards has picked up glutathione sound signature needed to throww off the virus.

    How do they get that much NAC in without side effects? I've read other places that supplementing more than 1200mg leads to some unhappy side effects.

      Yes, PAH is a real concern at high NAC dosing - so I'd max at 1800mg /d and glycine at 5g/d and be done with it. If it helps, it helps, if it's insufficient, so be it.

      @@kkostadinof According to Life Extension Foundation, the 2007 study that found NAC caused PAH in mice used the human equivalent of 20 grams a day. The 2010 LEF article also indicated they found no other evidence of NAC causing PAH and they questioned the findings of the 2007 study. Nevertheless, I think will try the 1800 mg strategy.

    It’s the overall wear and tear of worn out systems failing. Just a sign of a gray hair is a warning that there is progressive system failures mounting within the body due to aging.
    I had 4 mice: First one normal healthy aging and fed organic, distilled water, filtered air and lived just over normal life span but healthy to the end. Second one had supplementation geared for longevity, organic feed, distilled water, filtered air(read below). Third was targeted for genetic failures averted by specific supplementation, organic feed, distilled water, filtered air and lived a little longer than mouse one. Fourth one fed commercial off the shelf commercial feed, city water, unrestricted calories that had the shortest life, suffered arthritis, failing eyes, ears, very lethargic and mostly gray hair.
    RESULTS: Second mouse lived the longest, full energy, healthiest with twice the lifespan of all others with almost no gray hair. My theory is even if you target genetic failures as in the third mouse, the aging process starts to catch up to other systems in the body and the continuation of accumulated damage still occurs until death. Supplementing for longevity helps the overall organism to live the longest and healthiest as in the second mouse who suffered the least and enjoyed life the most. I’m on the second mouse diet!!!

      Great question. Eating more protein would be expected to increase glycine+cysteine intake, but as we all know, too much protein can shorten lifespan, at least in mice. So there's a fine line for that.

    Great videos ! Love the knowledge you impart , I have become an avid fan and am drinking in all your videos since discovering them. I have one question though concerning NAC and it is the concern of long term use of NAC and liver damage. I knew that NAC was used therapeutically for treating overdose with paracetamol but it was reported to have long term effects regarding not allowing the liver to regenerate. I am hoping I am wrong as I would like to start taking both glycine and NAC alongside one another in the amounts you have stressed ie 5 and 9 grams respectively. Any thoughts or opinions on this as I am keen to start this ( I would have taken oral glutathione, but I believe this is ineffective regards its efficacy).
