The DSLR has a small sensor which crops the image, compared to a full 35mm film frame. This is why DSLR lenses will normally be rated with a "35mm equivalent focal length" in place of true focal length. Nikon DLSR for example will accept old lenses from nikon 35mm film cameras, but when used on the digital camera it behaves like a longer focal length lens because the projected image is larger than the sensor and it gets cropped. To know the amount of the effect you must get the sensor size of your camera, there are more than 5 sensor sizes in common use. When camparing two sensors with the same megapixles, the larger sensor will have larger pixel elements that capture more light.

Yes, everything, starting from forceps, and media, to plates that you use in the process should be sterilized before putting them into a laminar flow hood.

What magnification is your microscope? I want to buy the same one but there are many options for magnification. 3.5x to 90x, or 7x to 45x. Amazon has one for $650:
