We seniors are counting on you and your collegues for a longer, healthier life.... and you're doing it! It's obvious to the lay-observer that longevity research has reached a critical mass. I'm hearing David Sinclair's words about treating aging as THE causal factor in nearly all common morbid diseases coming from labs all over the world now. Thank you so much for taking on this challenge!

I'm continually amazed at how you're able to explain the complex in a most accessible way, and with humor.

Brilliant explanation of the ISR and ISRIB! Cheers. And what a beautiful molecule: suppressing the high baseline ISR activity seen in ageing without affecting the spikes in the ISR needed to fight viruses etc.

I had spent 5 years studying yeast eIF2B (GCD6 and GCD7) and eIF2 (GCN3 or SUI2). It was a very interesting field. Thanks, Dr. Scheekey!

Excellent. Have been following aging issues for years (as a layman) and don't recall coming across ISR. Reminds me a bit of hormesis. Really appreciate the amount of work and level of detail which goes into your presentations. Thank you!

Your channel is very unique and interesting. Thank you for the time that you've invested in these videos and for your interest in making information about things like this available in a way that is easy to understand. You seem to have a lot of passion for educating. I wonder if being a teacher is in your future? Regardless of what you decide to do; I really hope that you will continue to produce content like this! Kind regards! Skyler R - BSW RSW, Canada. BC.

So, what about natural sources of ISRIB? Is this basically something that fasting can trigger? Are we talking about inhibiting mTor?

  • @biomoger_nature

Hexarelin, and si wu tang extract can also affects mTOR pathway according to my findings!

Last question, I'm not sure if you're familiar with Cerebrolysin, but if you are could you shed some light on whether prion disease, brain cancer, and other purported risks are serious concerns?

so can calorie restriction activate ISR through activating GCN2?

    • @TheSheekeyScienceShow

I was curious about this too - haven't managed to probably look into it yet... But as as i mentioned in the video, the ISR is an important process and although the stresses are "integrated" through the pathway it could be that inhibiting ISR when its induced by PERK (unfolded proteins) like in neurodegen is more beneficial than inhibiting ISR when induced by GCN2, but yeah, good question!!

Thanks, Eleanor! You make me wish I had a bigger brain … maybe I need ISRIB. The multiple layers of negative feedback (X inhibits Y which inhibits Z) seem like the submerged platform problem adapted for humans. I always feel smarter after one of your videos.
