I feel like that part that Dororo points at the eye (after he woke up from being unconcious) needed some more frames of him waking up, it felt weird you see him asleep and then insta woke and screaming, either way, great episode and great anime!

At this point, I think it's more likely that the father finds out that his first son is still alive (and is killing the demons) and will send people after him. Or if the father doesn't know about his son, he would want to kill whomever it is, regardless.
