He doesn’t guess. At 00:31 - 00:32 he stops and turns in monster form and you can see the canals symbol slightly to the right and up of the screen. You can also jump in this position to see it more clearly, but he can tell what it is from just the small portion shown. At 1:24-1:25 you can see him jump on to the roof and jump off. You can see the symbol as he is falling. Right before jumping he sets up the grapple on to the theater building so as he sees the symbol it grapples him back so he can open the door. Second player got the boxer district symbol. This one is easy, but after breaking the box with the belt, you jump on to the roof on the right and as you run across to go open the boxing ring door, you can see the symbol clearly on the left.
Also i THINK if I’m not mistaken… if you get a power up in that area the rocket launch initiates automatically. He popped reign drops which gave him drops initiating the launch. Crazy what you discover on this game 8 years later!

the last step is 4 player but you just have to kill the shadowman for most speedruns
