Thanks for the video. Have learnt some new things like playing the Bb 7 and its tritone substitution E7, one after another. I am trying it by leaving the Bb7 notes of D,F,G# after I have played it, and then just adding B note to the right hand to make it E7 b9 and playing it as an arpeggio. Also, I have learnt how to play the sus2 sus4 chord by combining the 4th and 5th chords.

How do we calculate the beat or timing of using it before the destination chord?

If you are playing 4 beats in a bar it's preferable to use the secondary dominant chord on the last beat which is the fourth beat before playing the destination chord. But you can also play it on any beat, it all depends on what you want to achieve.
