    As a recent breast cancer survivor, I’ve done a LOT of research on mitochondria and how to incorporate diet/lifestyle.supplement changes in order to stay in remission. Your videos so thoroughly explain the science behind so many of the claims I hear from other UA-camrs -who I generally avoid- but still occasionally watch. Thank you for your deep dive into the research papers which I often read but not sure I know how to interpret. Your lectures are always delightful, informative and charming! Keep up the awesome work!

    I have been fasting for my entire life. Not so much by choice. I was born with a very severe nerve system in my mouth and sinus. The nerves were so sensitive to everything, including smell, taste, heat and cold, and pressures. So from an early age I didn't eat much. If I did eat, it had to be soft and room temperature and I could only eat with either a plastic fork or a gold fork. By the time I was in high school, eating was just to much trouble. So I would only eat something at night just before bed. I kept this up until I was in my mid 50's when I had a accident and had to have all my teeth replaced with dentures. Holy crap!! I can eat anything with these teeth and have been, but it changed me. From my early 20's till I had the dentures put in I barely aged at all. When the family would go to funerals my sons felt bad when other people thought they were my uncles and dad. People think my younger brothers are my grand dads. But since I have been eating, the age is really starting to show. Besides not ageing very much at all, I never have been sick. Except for allergies.
    As for cancer? I know and have known lots of people over the years that survived terminal cancer thru extreme fasting. Not intermittent fasting. Intermittent fasting may slow things down a bit but as soon as you start eating again, things just go back to normal.

      Interesting story. I have read many stories like this and have been doing CR for decades myself. It 100% works. Especially, when you've been doing it for so long. You start noticing everyone in your age group looks much older. The strange thing is, less you eat, less sleep you need and the more energy you have. This sounds counterintuitive, but try it yourself. Low body fat is extremely healthy. We're just not used to it in the modern world. Ofcourse, one has to look out for eating disorders. With food it's simple, quality over quantity! Also, I once calculated and compared grocery bills to other people. Well, saved more then 2000$ yearly while feeling fantastic and never sick, eating staples like beans, lentils and oats. ;-).

      @@VeronicaMist Nutrient-dense foods. I'm a pescatarian myself. The trick is just to have 1-2 meals/day. These can be fairly large meals. Just water,tea or coffee in between. No-calorie beverages.

    I have been following Dr Mindy Pelz’s advice and it has really helped me. I have gone from being obese with associated health concerns to being a perfect weight for my height. She may not have the precision and in depth expertise that you do as a scientist - but she has a special ability to reach a non scientist population to massively improve their health; more than what could be said for many doctors.

      He claim may be from one of too many studies out there. Way to many to figure out wich ones are NOT from the big pharmaceutical companies trying to confuse and mislead people maybe even this that fasting could be bad in same cases, as more then half I have heard it is.

    Thank you for your calm clear appraisal of this influencer. The message for me is clear... fasting is relatively safe and probably the most effective.
    I would love to know more about which cancers benefit from autophagy and more information on how soon autophagy kicks in.

      for a 24 hour dry fasting, hgh appears around hour 13, autophagy at hour 16, intestinal stem cells at hour 24. but this is for advanced fasting. I don't know for water fasting.

    I caught her exaggerating positive results from a study about alternate day fasting. The study said people doing ADF lost an average of ~3kg a month - she said it was per week.

    Thank you! I am just a keen IF person and have been trying to find papers that said autophagy kicks in at 17 hours and couldn't find anything - most said anywhere from 24 hours and upwards and not to let it go on for too long either. So glad to have this query answered by someone who really knows

      from what I read autophagy kicks in at hour 16 of a 24 hour dry fast. hgh at hour 13. intestinal stem cells at hour 24 (per mit)

      @@miaya3898@alisoncross2399 But here's the thing: autophagy isn't binary. It's not "on" or "off." It's at varying levels of "on-ness." So, what level are they considering it "kicking in"? It'd be better to see a chart that shows the increase in markers for autophagy over time. There may be an acceleration of those markers at ~16 hours, but it's not a light switch that's flipped.
      I've been an OMADder at various times in the past, so this is something I'm keenly interested in. 🙂

    Why are so many of these UA-cam longevity specialist types chiropractors? Present themselves as a "Dr" which carries an implied expertise but then ends up they went to chiropractic school. Just something I've noticed. In contrast I'd say your expertise is well grounded. I won't be adding Dr Mindy to my subscribers list.

      Back in the day, chiropractors would give you advice on healthy eating and all the other stuff. They study the body in a different way, and expand their knowledge. As yourself, I don't care to see Dr. in front of their names, or at the very least they should tell us they are chiro doctors, or Bible doctors....

      Piperine, a compound found in black pepper at levels of ~2-7%, is a CYP3a4 inhibitor and also an inhibitor of the P-glycoprotein efflux pump. Here's a paper that talks about the bioavailability of curcumin in a variety of formulations: www.ncbi.nlm.nih.gov/pmc/articles/PMC6770259/
      Piperine is one of many options to improve bioavailability. In three different studies, the dose of piperine ranged from 5 mg to 24 mg to 20 *mg/kg*, which is quite a range. Even using the lowest dose, you'll need 100-200 mg of pepper. You'd probably be better off with a gram or more. Piperine's effect on curcumin metabolism is said to occur through inhibition of glucuronidation. Since glucuronidation is an important pathway for metabolism of xenobiotics, I'm not crazy about inhibiting it willy-nilly, not to mention the effects of 3A4 and the PGP efflux pump. I'd rather use one of the more sophisticated formulations of curcumin in order to avoid drug interaction effects.

    I’ve been on a low carb diet for about a year now and read or listened to all sorts of information about autophagy. While it’s an important part of physiology and shouldn’t be ignored, I have a problem with presenters who make it the be-all and end-all of metabolism. If we don’t overeat and leave time for our bodies to do jobs other than digestion, we will naturally cycle through its various processes, including autophagy. To put it simply, cells use energy to do the jobs they were meant to do, communicate with each other, create new cells and recycle their component parts when its time to do so. Each “job” takes place according to the cell’s environment at the time. Autophagy won’t take place while certain other things are happening. A high glucose environment prevents autophagy, according to much of my reading. As I see it, if the foods mentioned encourage autophagy, it’s because they provide something that is necessary to the process when autophagy is in progress. I would assume that they would not be providing glucose.
    Much of what I’ve read also seems to indicate that if a cell is constantly busy doing its primary job, it hasn’t the time to “clean up”. Waste products continue to accumulate and eventually cause damage.
    As we do know, a person concerned about whether this biologic process is occurring properly in his/her own body need only pay attention to eating real foods - not highly processed food-like substances - and allowing the body to be able to adequately rest from digestive processes, e.g. caloric restriction and/or fast.
    You were correct in that the amount of time necessary to induce autophagy hasn’t had enough research but according to the Cleveland Clinic, it appears that it reaches measurable levels somewhere between 24 and 48 hours. I’ve also read that it peaks at 72 hours, though I was unable to track down where I read that. (These were measures of fasting time.)
    Re: chiropractors outside their “areas of expertise”.
    I am unwilling to accuse anyone of being outside their area of expertise simply because the degree they hold or lack of one. Any intelligent person is capable of learning whatever they are interested in. I have 2 degrees and my husband has none. Yet he possesses more knowledge of history and geography than some of the professors I’ve known. I myself was passed over for a cardiac research position because I didn’t have a nursing degree. I was merely a certified cardiovascular tech with research experience in microbiology. I find judging people by paper achievements to be snobbish at best.

    I advise you to have a direct discussion of this science with the people you are constructively criticizing (like Dr Mindy). Im sure you could help fine tune their message, which has been very beneficial to tons of ordinary people who have struggled with poor health for decades.

    So regarding cancer and autophagy. How were the tumor enhancing effects of autopgagy in cancer cells demonstrated in the paper: in vitro / in vivo / outside of fasting? Did this show that fasting in some cancers increased tumor growth? Your video on cancer and fasting was superb but made reference to any cancer any promoting effects of fasting, that I recall.
    If so case that could be an important distinction - inducing autophagy outside of fasting, say, might be tumor promoting for some cancers in the presence of glucose but within-fasting autophagy could be tumor inhibiting with glycogen supplies depleted (say).

    it seems that the idea with MCTs more than olive oil (which has a lot of other stuff in it) is that it helps coffee raise metabolism especially lipid metabolism, sort of priming a physiological energy pump without initiating protein synthesis, and with that initiating conditions for autophagy perhaps with challenges like climbing stairs. Daily, this helps to burn fat and with the body in this pre-phagy state, autophagy is potentiated.

    I recommend checking out Thomas DeLauer on UA-cam. He does “healthy keto” and he always references studies. He’s also weary of saturated fats and tries to primarily consume the “healthy” oils like olive oil, and avocado oil. 5:00 even more importantly although I can’t remember off the top of my head, I’m betting his videos on fasting have a lot more data and studies backing them up.

    A fews seconds fasting does get rid of damaged/metabolise cells, even though as you point it isn't autophagy. Longo made this observation on white bloods. During fasting white blood cells were depleted, but then receovered above baseline - this way fasting can rebuild a damaged immune system, such as though chemo - he claimed.

      Well, as Nick pointed out its not autophagy but depleting white blood cells isn't a good thing for the immune system - except in the context descrbied where there is a plan in fasting driven through necessity: there is no energy coming in so something must be metabolised. But you wouldn't want to take something that destroys your white blood cell count - this is why Longo recommended fasting for post chemo - when those white blood cell levels are low and so can be rebuilt.

    Another very good and informative analysis. I didn't know that curcumin indirectly reduced mTOR activity -- does that mean frequent ingestion in turmeric or supplements is likely to suppress muscle synthesis, or would the effect be too small?

      That’s the question that immediately leapt to my mind too. I doubt there are any good RCTs on whether cucumin’s mTOR inhibiting effects reduce muscle hypertrophy but it would be an interesting thing to study!

    Its interesting there is so much criticism toward chiropractors. There is tons of science behind it as well as acupuncture. Both of theae treatments saved my great danes life when she suddenly was unable to walk. All the conventional vets couldnt help. She was on muscle relaxers, gabapentin, tramadol and remadyl (prescription strength anti inflammatory for dogs.) None of the meds helped. One chiropractic adjustment and acupuncture and shes up walking again. (Not perfect yet but she was NOT able to stand or walk for 3 weeks with regular vet interventions.) In most households she would have been put to death. I thank God for an educated, good chiropractor and acupuncturist.

    Quite the opposite experience here.
    Fasting or eating very little just made me look older.
    On top of that I was always lethargic and hungry.
    Sensible eating and caloric restriction is what worked for me.
    People will be affected differently.

      I have pretty good results with fasting but only up to 48 hours any longer and it has the opposite effect for me too

    9:02 I understand your thought process and I wish the data was better. I would like to point out that if 2 out of 3 proteins are elevated, and as you say “autophagy is starting but not completing”, perhaps EVOO in combination with other factors will still act as a booster because it is increasing some of the necessary proteins.

    Very good emphasis! I appreciate both types of focus. The science of such COMPLEX systems always leaves a LOT more to be desired toward quality assurance and informed consent, THAT is the scientific method. The more I know, the more I know I don't know, ya know? We are desperately in need of better food sourcing methods than the current status and trends of western dietary practices on such a massive scale. We desperately need more food science, and applied science guiding 'real food' quality (and quantity) going forward. The current status of the western medical paradigm is very good for acute medical science, but IMHO we are falling flat on our faces for chronic disease root cause assessment.... because of (mostly) 'profit at any cost' marketing BS, which leads us FAR away from science, and from essential funding of science too. We now appear to be in DEEP trouble (fat, and insulin resistant, and beyond) well into ill health for vast numbers of us. Both of you are doing great service toward helping, and both are synergistic toward getting a better handle on it!
    Thank you both! We need both approaches, speaking two languages toward influencing wellness toward better paths forward. These methods are NOT mutually exclusive, but synergistic.

    Great video on proven details of autophagy. You agreed coffee was good and some smaller effect for autophagy with mushrooms. But with both can’t combine with other foods as then these other foods most likely to turn on mtor so then no autophagy. So seems then best to have in morning with overnight fast just with the coffee and mushrooms. A product with both is Four Sigmatic ground coffee mix. Too as you mention autophagy is internal and hear/see many saying incorrectly that is for getting ride of damaged cells. When is i think strictly rebuilding inside cell certain damaged organelles.

    I've been meaning to ask. What do you think of autophagic flux? Some say just looking at an increase of lc3-II isn't enough and looking at -/+ autophagosome nhibitors is better to see how much is broken down over time.

    Noooiice Video! I hope she's not mad at you for acting like a "state of the fact - correction - drone". UA-cam gave us the opportunity to transport knowledge out from the bubble of where it got researched, to people like me, which are not deep diving science heroes. And this happens by people like her, trying to understand complex systems and their result for our body and health. But these "influencers" may also lack of in depth knowledge about such very numerical data analysis, the result and its meaning. Thus not fully understand the process behind the word in its wholeness, at least to actual level of research. So now "influencers" like you jump in and bring up that nerdy deep factual and empirical knowledge (omg I love it so much), which is not the kind of information a normal person, without a degree in such a field, understands. So while she influences the wider broad population, you certainly influence me, as a nerd, other nerds and I hope such "influencers" like her which attract the broad population. So she can use the knowledge you publicize here on UA-cam, to transform it into easier-understandable information packages, which then reaches people that are trying to be healthy but not having time and interest in studying biology etc. That's Awesome! A system of transporting knowledge, to spread knowledge about a system in biology. UA-cam and Creators like you Physionic make knowledge a lot more accessible and understandable. Thanks for all that

    I just subscribed to your channel. I thought when I first began to watch this that you would just be negative but you are truly unbias and data based. I do low carb cycling, intermittent fasting and longer fast. I have not made it to a 3 day fast yet. I have seen some health benefit to this in firstly losing the pandemic weight I gained. I started having a problem with my blood sugars and heart well they are on the heal with this program of moving between the two energy systems of the body- fat burning and sugar burning. I find this works best for me. I did try Keto for 45 days and I ended up gaining weight. It suppressed my Thyroid.

    If you use curcumin (or tumeric) be sure to combine it with a little black pepper. As Nick points out in another video, pepper increases the absorption of curcumin twenty-fold.

    @physionic How do you heal from a vaccine injury ? Mitochondrial autophagy is key to this. I am 2.5 years injured and I seem to be going downhill. My brain does not work anymore like it did. I was a molecular bio student.

    I think mTOR is the key to activating or enabling autophagy. Dr. David Sinclair explains that mTOR turns on when a certain level of protein is achieved. This enabling point has to do with the body switching from a survival mode, not enough or barely enough protein, to a growth mode. In growth mode autophagy is not needed because there is an abundance of protein. This is my interpretation of what Dr. Sinclair explains.

    Have you done a study on Chanca Piedra and the chemical results? Not too many spek about this study. I CAN truly testify that it does wonders for symptoms of PKD. I also experienced COMPLETE CELL REGENERATION from a recluse bite using turmeric, cayenne, ginger and pepper for a drink and used EVOO and the same in a paste in my wound. Today, it was NEVER there, and my DR. swore he never seen it (though he cleaned it and dressed it BEFORE i went to holistic and ayurvedic healing) .
    If you could give me the scientific breakdown of Chanca Piedra and how i can get the most out of it. Ty

    The idea behind the MCT oil and olive oil which she doesn't go into in. Is that it's used to keep people in a ketosis state. It's ketosis that helps autophagy not the oil themselves. So if you had lots of carbs there would be no autophagy.

    Thanks for all your awesome work. It’s been 6 days since my water fast, although my prep prior to the fast was not great (2 Filet o Fish, some fries) I’ve drank a mixture of Magnesium Citrate, Sodium, Potassium Chloride. And alkaline spring water. I’ve gone from 191 to 179 Lbs as a 24 y/o 5’7

    Curious - What is it called when one cleans up dead protein from a normal lifestyle, harder work etc. - injury etc or heavy sports/training. Where the body can't clean up the damaged or dead muscle tissue? Back in the day this was called DOMS I believe. Bonus question - Is this not any part of Autophagy? Many thanks and 👍 as usual
