Is it that although the amplicon size must not differ too much it must still be different enough so that distinct bands may be observed during gel electrophoresis?

Awesome 3 minutes . Thanks a lot. each of my samples contains one bacteria and one virus. Can I use multiplex pcr to quantify them in each sample and compare those ?
@@dr.urmibhattacharjee6919 yes you can, if the pcr product size is different lets say 400bp for bacteria specific gene and 600bp for virus specific gene then its possible .

I have a doubt sir, if the 3 different primer set have slightly different annealing temperature say for eg: 53.1;53.7;53.5 which one should i choose or should it be exactly same annealing temp for all primer sets?

Not mandatory It depends on what you want to detect, depends on your target's availability. As he told the criteria for multiplex, it must be ensured first before having targets.

Il est nécessaire d'ajouter l'ADN du virus dans un autre tube et faire le même PCR pour le positive contrôle
Viral DNA is already present in the patient's sample. No need to add it separately.
So for covid we can use multiplex PCR??
