      Progressive staining is a slower staining process in which the tissue is left in the staining solution just long enough to reach the desired endpoint. In contrast, regressive staining is a more rapid staining process in which the tissue is deliberately over stained and then de-stained.

      Eosin is very soluble in water and diluted ethanols. So if you do first Eosin staining and then go through aequous, acid hemalaun (pH = about 3), then bluing for a few minutes, no Eosin-staining would be left. It is the same, as if you let your already stained slides sit too long in the rinsing water and dehydrating ethanols. Eosin will be gone.
      Try it.

    How much time is required to prepare H and E slide for reporting?

      Here is my protocol. It works in paraffinsections (5-10µm) as well as in mounted frozen sections (10-30 µm). Dewaxed and defatted sections rinse in destilld water.
      1. Mayer's Haematoxyline (0.1%) - 5-10 min; 2. rinse well in destilled water; 3. blue in running tape water for 10 min., 4. 1% alcoholic Eosin- solution 1-5 min,; 5. rinse in destiiled water; 6.diffenciate in 70% ETOH (check under the microscope) and 2x 100% ETOH (10 dipps each alcohol); 7. Isopropanol 2 min; 2 x Xylene 5-10 min each; 8. mount in Depex or another xylene soluble mounting medium. Start with a test slide to optimize your stainig times an control after every stainig step the quality under the microscope.

    How to prepare percentage of alcohol,like 90%,80%,70%....etc mam let me know the method correctly..

      70%alcohol - Suppose To prepare 100ml of 70% alcohol .
      Add absolute alcohol (100% alcohol)
      70ml & add 30ml of Distilled water.
      (70% absolute alcohol + 30ml distilled water.)

      Hydration means addition of water . you have to hydrate the tissues so that they can absorb stain. Easily.
