Very useful framework. I might add that the simplest models of single cells metastasizing may be too simple. Many metastases seem to be initiated by clusters of cooperating cells including macrophages. I like this finding, because macrophages are professional migratory cells rich in the ability to dissolve intercellular barriers, crawl into and out of blood vessels, and support new growth (at the new site). This is still under intense study. Thank you for mentioning side effects of current checkpoint therapy. More research!!
