I can help add a little more context with what's going on with the cards in the hand to go with the body language. Let's start with the flop which is the first three community cards placed in the middle. The first "red" flag we see is the flop is all hearts. The odds of flopping a flush if a player is holding two cards of the same suit is 118 to 1, or less than 1%. LeChiffe is a mathematical, statistician player so he knows those odds. His pair of 2's won't hold up to take the pot so he bets here hoping to represent that he does have a flush. This is where his duress starts and the tell appears. Bond calls his bet. If LeChiffe represents the mathematical player, then Bond represents the people player. LeChiffe plays the cards, Bond plays the player. Bond has deduced that LeChiffe's bet is weak. We never see Bond's hand, put I suspect he's either made a pair with the flop or has the Ace of hearts drawing for the best possible flush (in poker terms, the nuts).
The fourth card is another 9. This pairs the board which makes a full house now possible which beats a flush. LeChiffe bets again now with two pair, but now Bond is distracted by Vespa. This is critical key moment in this hand. LeChiffe doesn't want to check here as to do so would be losing the initiative and practically yielding the pot to Bond. His bet however is offhandedly called by James 'distracted by the sexy' Bond.
The fifth card is the 2 of hearts. This gives LeChiffe the full house. Indeed, both players have now relaxed their posture (you said James, but did you mean LeChiffe?). I now have to put Bond on the nut flush and so does LeChiffe. Somewhat agreeing with your analysis about Bond's forced smile after the hand is over that further points to Bond having the flush. I also read into that smile as Bond's way of saying "Yeah, you won that hand, but I've got your number now."
There is a chance that Bond has absolutely nothing and is only calling LeChiffe's bets to either find out if he's right about the tell (he is) and to antagonize LeChiffe, but I doubt he'd be that reckless. Then again, this IS Bond we're talking about here.

I just realized that the reason Le Chiffre put his finger to his temple was because the vein that runs through his temple and under his eye was bulging. This was caused by his injury, so when blood starts to pump more, either from excitement or frustration, the vein becomes more noticeable, and he has to press his finger on the temple to keep it from giving him away.

As you can see enigma has his hands together on the prop table, which is a self soothing action he also looks directly at the stressor, the camera, meaning he is focused and this can be seen by him leaning toward the stressor. @@Samanosuke1138
Disagree, but if that's what you believe that's fine.
But it was Vesper who betrayed Bond. Not Mathis. She knew about the "tell", because Mathis explained it to her whilst watching the poker game. She was an unwilling double agent, she didn't want anyone to get hurt, but her mission was to make sure Bond lost the game. That's why she did her best to distract him at the table and refused to bank roll him after he lost.
Le Chiffre using her to cause the crash is all about him using what he knows to be Bond's weakness, his blind-spot, against him. It was all manipulation. He placed her in danger and used the idea of threatening her in interrogation scene because he thought he could manipulate the code out of Bond. Plus she had failed in her mission and was next to useless to him at that point.
And as for Mathis, it's even confirmed in the next film he was innocent and was compensated by MI6 with the villa where he is now retired. The whole reason they agree to work together again is because Bond knows he is the last person he can trust.
But I guess that's my interpretation.
In short, I think Le Chiffre is a great villain as he uses manipulation to get what he wants. Just like a great poker player, he doesn't play the cards, he plays the person across the table from him. He used every trick in the book to try and beat Bond but in the end he is killed because he could not break him.

As an autistic person, one of my first body language lessons was how to read "tells" in poker. Leaning forward meant bluffing and a bad hand. Leaning back meant relaxation and a winning hand. My second, as a cartoonist, was superhero and supervillain character designs in comics. the former tend to face the reader, smile a lot, and have open stances, often with arms akimbo. Their costumes tend to be red, yellow, and blue. Supervillains tend to frown and scowl often, stand or sit sideways, facing away from the audience, hand-wringing, or rubbing and have predatory, sneaky body language, such as claw-like hand movements, walking on tip-toe, etc. They tend to wear black, red, blue, green, and purple costumes, especially spiky black armor and black and red opera capes with high collars or hoods. Skulls are a common chest symbol or mask. My third, as a martial artist, was both how to spot and avoid "telegraphing," a common boxing error, which gives away one's next move. Pulling back a fist or foot means one is about to strike with that limb. Leaning forward with arms wide open means a potential grappling attack.

I wish he did the whole scene james gets so mad he even tried to kill him until he’s stopped by the cia agent played by Jeffrey Wright who is seen at the table with his hands open showing he’s probably not scared and confident because he’s used to that stress as an agent. He gives james the money to buy back in and eventually James gets the upper hand but madds was playing him the whole time in the beginning and is winning the game until james can buy back in. It’s a real shame madds wasn’t as big as he was when this came out cuz it would be awesome to see him do more in another villain role but he was amazing in this as well!

So have you seen Madds in Hannibal? You won't be disappointed. Gillian Andersen is excellent in this -- as she is with Jamie Dornan in The Fall. Another must see......perhaps Enigma, you could do the interrogation scene from The Fall in the final episode. Soooo Sublte but telling!!!

I’ve seen Hannibal and it’s pretty much my fave show that’s how I’ve learned to love mads. They canceled that too soon real shame.. I haven’t watched the fall but my fiancé really liked it.
