To get the meaning and emotion that this song truly carries you really need to see the play (it is available on Disney+, not sure what the availability is in the UK). The women here are the actresses who played Eliza in all the subsequent companies after the original cast stepped down. It is an expanded rework of a song Eliza does in the show called 'Burn'.

It can definitely be confusing if you haven't seen the show, but the whole song is sung by just one character: Hamilton's wife Eliza (The five different singers are all actresses that have played her on stage) who is devastated after Hamilton makes the details of an affair he had public. This is an early version of the song. The "I see how you look at my sister" line (which is my absolute favorite but is not in the musical version) is a reference to the extremely close relationship Hamilton had with Eliza's sister Angelica. That's not who he cheated with, but it's a theme in the show and even historically accurate that they wrote letters to each other and held a much deeper affection than is appropriate for a brother in law or sister in law.
*I know I'm late to the video here so you may have gotten this cleared up already or even seen the whole musical but I thought I'd mention just in case!
