You can, but the idea is that the queenright finisher spends more time feeding the cells as they aren't producing queens under the emergency response. That being said I have had some really good queens come from a queenless starter/finisher. Just make sure they are densely packed with bees and you'll get yourself some good cells. The other advantage to separating the starter and finisher jobs is the ability to put a new graft into a starter every other day. In the case of this video I think I ran three grafts through that starter before allowing them to keep a cell and become queenright.
