This isn't his mother and sister's fault. Mustafa was actually supposed to die, his brother Ahmed I spared his life and ended fratricide. All Şehzade who were eligible for the throne lived in the Kafes from then on. He could never have governed his own province, by that time the princes weren't allowed to do that, instead they were confined in the Twin Kiosks/Crown Prince Apartments.

Everyone keeps saying kosem lost her innocence. I wonder how many of us can keep up with innocence in a system where you will end up dead if your guard is down. Sure kosem became ruthless over the age but she never wanted all this to begin with. Unlike other women, even when she was brought to the palace she never wanted to be in the harem or seduce the sultan despite Ahmet's interest. She finally settled down when she realised she is stuck for life. Ahmet left the entire kingdom and the life of his kids in her hands when he died. If she treasured her innocence everyone she loved would have been dead the very next day. She became what she was because the situation demanded. She ruled the kingdom as regent for many years. Once you taste power and in her case she ruled the entire empire it's not easy to give up everything and get ruled. No human likes to be ruled or be a subordinate. None of her kids were capable enough to rule the kingdom as far as they show in the show. Therefore given all these circumstances she had to give up her cherished innocence.
Mustafa pinpointed exactly who has she become. Now I would say as Safiye did not love her grandson she loved her son Iskender so much that after his death she decided to follow him. Kosem even now plots against her own son because her dearest love is a throne , not a person who sits on it.

Till the tenure of osman kosem had to live in Old Palace. Until she plotted against osman and killed him. Then again mustafa got the throne and she deposed him and made murad to take the throne.
