Yes For any ISL switch port mode should be E.

Disabling port or a port channel is persistent ? I mean if I disabled Port or a port channel and reboot the switch, port or port channel should remain disabled until I enable it like how we use persistentdisabled/persistentenable in brocade switches.
The port or port-channel will only remain in an "admin down" after reload if the config was written. Whatever the port config states in the startup config is how the port will act once the switch is reloaded. Try this command to check the start-up config:
show start-up | i
