The polyglutamine diseases "more apt to cause damage with every generation." Is this generation referring to generations of individuals, or generations of fibril development?

She means the generation of individuals. This group of neurodegenerative pathologies like Huntington is known as autosomal dominant disorder, so we expect to see that 50% of child will be affected (assuming one of the parents is normal who doesn't carry the mutant gene). She also means that as the mutation is passed from one to the next generations, the disease appears in children earlier than their parents, gets severe in terms of clinical symptoms, which caused by the increased number of poly-glu repeats and genomic instability.
