Defining neighbours is done based on the sensor positions of the specific montage you used. FieldTrip (but other toolboxes as well) for example has a number of such templates for certain common systems. If your setup is not included in these, you can create this neighbour definition yourself for example through 2D triangulation of the 3D positions of your channels, which should be known.
