We had to read it in high school in German class and it's the type of book you can torture teenagers with, because it's so boring. Effi's main antagonists are societal conventions and the feeling that the house she moved into is haunted. She has a tragic ending. Therefor, a suitable world for that combination would simply be 19th century Prussia / German Empire.
That thing with the Neanderthal genes... You must be careful with that, because people with the lowest Neanderthal admixture are all sub-saharan Africans, while Europeans and Asians all have Neanderthal and/or Denisovan admixture (and possibly more). So, depending on how you construct it, it might come across as vilifying a certain race.
