Hi. Just one thing, the colorimetric method if IHC depends upon an enzyme and not a substrate being conjugated to the primary or secondary antibody. The substrate (DAB or FAST RED for example) being added in subsequent steps to produce localised (brown or red) coloured precipitate for visualisation
