Then it's very probably that you don't have the same concentrations values for your samples, in that case you can calculate the ic50 separately for each sample

Hi thanks for your comment The point (0,0) is a known point because in my case there is no absorbance without concentration, this will help you with the regression

Also what if I tested my sample at different concentration and none of the concentrations inhibit 50% does that mean I wont be able to calculate the IC50 or it just means I will have an extremely high IC50 value?
hello thanks for your comment, the IC50 is a concentration not a percentage, IC50 means the concentration value that inhibit 50% of the population so you can't do it without concentrations values. if you tested your samples at different concentrations and none of them inhibit 50% that mean you must prepare high concentrations to get the IC50, if you do that and you still having the same result that mean your sample have no effect

Hi, I checked the antityrosinase activity of my sample at 200, 400, 600, 800 at 1000 μg/ml and the results were 1.25, 3.925, 5.775, 6.625 and 8.75 respectively. I used kojic acid as standard at the same concentrations and got 16.437, 24.625, 32.625, 47.5625 and 86.75. I used Y=mX+C to calculate the IC50 values and I got that of my sample to be 0.2667μg/ml and kojic acid to be 702.55μg/ml. Please I need someone to use graphpad prism to validate my result 🙏
Trust me best vedio on using Prism for IC50 calculations, highly appreciate your efforts.

@@sanjamatic8767 You need test MRC-5 in those doses that can kill from 0% to 100%. Now when you perform the calculation you can get the IC50.
