Selection and Characterization of hSTING-Agonists via in silico modeling and SPR

  • In this study, Cayman Chemical performed our established computer-aided screening techniques using Schrodinger to select hSTING agonists for further characterization via Surface Plasmon Resonance (SPR). We established a SPR platform to evaluate binding kinetics and affinity measurements for the agonists. Synthesized agonists such as canonical CDN (2’,3’ cGAMP), amidobenzimidazoles (diABZI agonist-3), and purified recombinant hSTING variants were used for SPR analysis using a Biacore instrument. Here we report the selected agonists affinities was improved compared to previously reported values. These findings could provide guidelines for the design and development of hSTING agonists research.
