The ModPodge acts as a resist similar to shellac, but it is waterbased so clean up is easier. If you wanted color, you would have to do the underglaze first, then use the resist on top. If you put the underglaze over the resist it will not adhere to the clay.
It has no adverse affect on the clay. It's a small amount of resist on per with waxing the bottom of a pot. Which btw, you could use wax resist instead but it doesn't adhere as well and sometimes easily peels away.

Could you use those tiles as stamps of they are fired?
Do you mean as bisque stamps on wet clay? Then yes! You would want the etching to be somewhat deep so the image transfers well.
