  • This video shows how I make a peripheral blood smear and how to perform a manual Wright Giemsa staining procedure on batches of 10 slides. Please make sure to label your slides with pencil or a chemical resistant marker. Pencil is the best choice.

    Quick question.. my protocol says to use absolute methanol and fix for 5min… is 5min for fixing really necessary? Also, do you have to stain the cells immediately after fixing? Can I for example wait up to 1hr after fixing to stain my slide?

      Great questions! I would say to follow your protocol on the timing. Under fixing a slide can really be a problem. As for staining immediately after fixing, I think you can wait. If you are waiting, make sure the slides are air drying and not sitting in a solution or on a heater.

      A fixative is a solution that maintains cellular morphology and prevents cellular degeneration. Some fixatives may also be used to adhere a specimen to the slide.

    Question... How much of the stain do you pour in the chamber?
    I'll be doing this technique in a couple of days.

      I pour up to the notch where the chamber widens at the top. The stain and alcohol evaporate really quickly. So, either weigh the lid down in between uses or keep refilling. Hope you have a great time!
