Great info, thanks. However I really need some help. I have a brand new trinocular stereo microscope and I want to use my Canon 70D to get some shots but they are too dark to see well. Because the adapter that is used you cannot attach a lens so the only adjustment you get is shutter speed and ISO. All focusing is done with the microscope. Do you have any suggestion or know anyone with this set up.

You should do a video about yourself and your background.

You're not using this kind of gear properly. Just buy an automated focus rail and do serious stackings to get any amazing results, this magnification is not for hand-held shooting.
To be honest hand-held staking works OK-ish only for 2:1 magnification ratio maximum. This gear here is intended for extreme macro.
